黄河流域青虾遗传多样性分析

【摘要】 本文采用20个微卫星分子标记对黄河流域7个青虾群体进行了遗传多样性分析,采样点包括东营,济南,东明,济源,三门峡,偏关,银川。结果表明:7个青虾群体平均等位基因数(A)为4.2643,平均有效等位基因数(Ne)为2.6682;20个位点平均多态信息含量PIC为0.5575;7个群体期望杂合度平均值为0.5609,研究结果表明黄河青虾遗传多样性丰富。分子方差分析(AMOVA)结果显示,黄河青虾7个群体间6%遗传变异来自群体间,94%来自群体内,两两群体间Fst值在0.0172-0.0791之间,表明群体间已明显出现分化(P<0.05),但分化程度中等。Hardy-Weinberg平衡检验表明,7群体中有多个位点偏离Hardy-Weinberg平衡。各群体间遗传距离在0.0455-0.2223之间,UPGMA聚类分析表明,7个青虾群体可以分为2大类。黄河山东段的东营,东明群体最先聚为一起,然后与济南群体聚为一起,最后与黄河河南段的三门峡,济源群体聚为一大类。黄河山西段的偏关群体与黄河宁夏段的银川群体聚为另一大类。

【Abstract】 Twenty polymorphic microsatellite markers were applied to investigate the genetic diversity of 7 Macrobrachium nipponense populations in the Yellow River basin.The average number of alleles(A) and effective numbers of alleles(Ne) in 7 populations were 4.2643 and 2.6682 respectively.The mean PIC value of 20 microsatellite loci was 0.5575.The average of expected heterozygosity(He) of 7 populations was 0.5609,which showed genetic diversity of the Yellow River Macrobrachium nipponense was rich.The analysis of molecular variance(AMOVA) indicated that almost majority of the variance in the Macrobrachium nipponense was within populations(94%),and 6%was among populations.The Fst values between populations was 0.0172- 0.0791(P<0.05),Which showed the genetic divergence between populations in the Yellow River was intermediate.Hardy-Weinberg equilibrium analysis indicated that deviations from Hardy-Weinberg equilibrium existed in all 7 populations.The genetic distances among populations ranged from 0.0455 to 0.2223.The UPGMA tree showed that PG and YC stocks formed one independent clade and the other 5 populations formed another one.
