棕点石斑鱼雌核发育的初步研究

【摘要】 本实验采用灭活的鞍带石斑鱼精子诱导棕点石斑鱼卵子进行雌核发育:使用不同剂量(1090 lux,720 lux,602 lux,438 lux)的紫外光分别处理鞍带石斑鱼的精子,在未加倍的情形下,照度为1090lux和720 lux的处理组均未发生卵裂,438 lux及602 lux的处理组胚胎卵裂正常,但表现明显的发育畸形和典型的单倍体综合征,并在孵化前后全部死亡。采用冷休克法加倍卵子,分别在授精后3min、4min、5min和6min对激活卵子进行6℃冷休克处理,结果显示:3min组受精卵不能正常卵裂,5min组大部分胚胎在神经胚阶段死亡,4min组和6min组受精率分别为24.7%和58.6%,分别有17.1%和20.2%发育正常的胚胎孵化出膜。

【Abstract】 Using irradiated sperm of Epinephelus lanceolatus induced gynogenesis in Epinephelus fuscoguttatus.The sperm of Epinephelus lanceolatus was irradiated by different illumination which was 1090 lux,720 lux,602 lux and 438 lux.The results showed that the egg samples of 1090 lux and 720 lux were not cleavage.The rest of the two samples appeared normal cleavage but presented developmental malformation and typical haploid syndrome in later periods and all fry died during the incubation.Activated eggs were handled with 6℃cold shock at 3min, 4min,5min and 6min after insemination.The results showed that the 3min sample presented abnormal cleavage.Most of the embryos in 5min sample died in the age of neurula.While the rate of fertilization in 4 min and 6 min samples respectively was 24.7%and 58.6%and the normal embryos hatchability respectively was 17.1%and 20.2%.
