中国自然四倍体泥鳅染色体减数分裂的分子细胞遗传学观察

【摘要】 为探讨自然四倍体泥鳅的起源及形成机制,对成熟的卵母细胞和初级精母细胞的减数分裂行为进行了分子细胞遗传学观察。结果表明,四倍体泥鳅中无论雌、雄均发现0-6个四价染色体和38-50个二价染色体。四价染色体数个体间无差异,而个体内不同细胞存在一定的差异。并且在精巢中观察到由于染色体数目加倍而形成的高倍数体,和由于分裂异常形成的非整倍体。同时,在四倍体中,2个二价染色体的端部观察到FISH信号和CMA3阳性部位,而这些在四价染色体上没有观察到。从上述结果可知,我国自然四倍体泥鳅起源于同源四倍体,目前,正在二倍化过程,但二倍化未完全。
【Abstract】 for study the origin of natural tetraploid loach,meiotic figures using natural tetraploid loaches collected from Yangtze River of the germinal vesicles of mature oocytes isolated from the ovary were analyzed.The results showed that several(0 to 6) quadrivalents and a large number(38 to 50) of bivalents were observed both in male and female tetraploid loach.The number of quadrivalents was the same among individuals,but different among cells within one individual. Hyperploid due to the doubling of chromosome and anueploid due to the abnormal meiosis were also observed in testes.FISH signals and CMA3-positive sites were also observed in the terminal of two bivalaents,which were not found on quadrivalents.Therefor,it was suggested that,the natural tetraploid loach is could be autotetraploid in origin,and the subsequent rediploidization has not been finished yet.
