杂色鲍皮肤桥蛋白克隆及其病原感染后的响应表达

【摘要】 尽管贝类的皮肤桥蛋白是一种具有多种功能的基质蛋白,但是对其功能研究很少。本文报道了一种贝类的皮肤桥基因——saDPT,克隆自腹足类杂色鲍,命名为saDPT2。其全长cDNA序列为620bp,开发阅读框为53 1bp,编码177个氨基酸。其推导蛋白的预测分子量为20.7kDa,等电点为4.62。同大西洋鲑的血细胞凝集因子具有最高的一致性(43%)。生物信息学分析表明saDPT2的推导蛋白具有皮肤桥蛋白保守基序,即含有10个氨基酸残基的S-X-H-X-N-X-Y-E-D-R基序。该基序类似于哺乳类6个氨基酸残基D-R-E/Q-W-X-F/Y。利用实时定量PCR检测了鲍各组织中和细菌感染后的表达情况。结果表明在检测的组织中都有saDPT-2基因的表达,其中血淋巴中表达量最高。弧菌感染后saDPT-2基因显著上调表达。这些结果表明saDPT-2基因对细菌感染有响应,在免疫系统中发挥了一定作用。

【Abstract】 The molluscan dermatopontin(DPT) has more general functions than matrix protein, but there is little knowledge available.In this study,a molluscan dermatopontin gene,saDPT,was cloned from small abalone Haliotis diversicolor.DPT-2 full-length cDNA sequence was 620 bp, with a 531 bp open reading frame encoding a protein of 177 aa.The molecular mass of the deduced protein was approximately 20.7kDa with an estimated pI of 4.62,and highest identity (43%) to Atlantic salmon Salmo salar hemagglutinin/amebocyte aggregation factor.Amino acid sequence analysis revealed that saDPT-2 shares conserved signature motifs with other DPT proteins,including three repeats of 10-residue motif(S-X-H-X-N-X-Y-E-D-R),which is similar to mammalia 6-residue repeating sequence of D-R-E/Q-W-X-F/Y.Quantitative real-time PCR was employed to investigate the tissue distribution of saDPT-2 mRNA,and its expression in abalone under bacteria challenge.The saDPT-2 mRNA could be detected in all examined tissues,with the highest expression level in hemocytes,and was up-regulated in hemocytes after bacteria injection. The results indicate that the saDPT-2 play a role in abalone immune system.
