三角帆蚌钙调素及其类似蛋白的基因克隆和表达分析

【摘要】 钙的代谢是贝类的一个重要而复杂的生理活动过程,它受到许多调节因子的控制。其中钙调素及其类似蛋白被认为发挥了关键作用。我们采用SMARTer RACE技术成功克隆了淡水育珠贝三角帆蚌钙调素(CaM)、钙调素类似蛋白(CaLP)cDNA全序列,并对两者mRNA组织表达量进行了荧光定量PCR分析。结果表明:CaM和CaLP在编码区核苷酸相似度为79.6%,CaLP比CaM具有更长的3’UTR序列(分别为716bp和158bp);CaM和CaLP在组织表达上存在明显的差异,1-3龄蚌中CaM在鳃、外套膜、腹足中表达量均较高,CaLP只在外套膜中表达量高。因此,我们推测CaM和CaLP可能在三角帆蚌钙代谢中扮演不同的角色。
【Abstract】 Calcium metabolism in mussel is a very complicated and important physiological and biochemical process,which is regulated in many regulatory factors.Among them,calmodulin (CaM) and calmodulin-like protein(CaLP),are thought to play a key role in the calcium metabolism.In this study,a full-length CaM and CaLP cDNA were isolated from the freshwater pearl mussel(Hypnosis cumingii) using rapid amplification of cDNA ends(RACE) approach. Comparing to CaM cDNA,CaLP cDNA has a long 3’-untranslated region,which are 716bp and 158bp respectably.Real time PCR analysis showed that CaM mRNA and CaLP mRNA had obviously different expression models in mussel tissues of 1-3 years.CaM mRNA was constitutively expressed in the gill,mantle,foot,while CaLP mRNA were only highly expressed in mantle.In summary,our results suggest that the CaM and CaLP might play a different role in the regulation of freshwater pearl mussel calcium metabolism.
