【摘要】 目的 :对地瓜参总皂苷进行溶血性试验和急性毒理试验,为其进一步研究和利用提供依据。方法 :以纯度为41.7%的地瓜参总皂苷进行活性测定,采用"溶血与凝聚检测法"对地瓜参总皂苷的溶血性进行试验,采用《GB 15193.3-2003急性毒性试验》进行毒理试验。结果 :地瓜参总皂苷在终浓度为0.2 mg/ml时120 min内出现部分溶血,而180min时显示全溶血。实验各浓度的溶血率均>5%。在最大耐受剂量(Maximum tolerated dose,MTD)为8000mg/kg时,小鼠空腹经口灌胃连续观察14 d未发现任何异常。结论 :地瓜参总皂苷的溶血性很强,在较低的浓度条件下都能产生溶血,这可能与其皂苷结构有关,其原理可能是地瓜参皂苷能与兔红细胞膜相结合,导致渗透压发生变化而发生裂解,造成溶血现象。地瓜参总皂苷口服无毒可能是因为经过动物胃肠道的消化分解后,皂苷被水解成皂苷元及其他产物而结构发生变化,被动物吸收后未发生毒副作用。急性毒理实验表明地瓜参总皂苷在一定范围内属于实际无毒。更多还原

【Abstract】 Objective:Did the hemolytic test and acute toxicological test to provide the basis for further research and usage of total saponins from Acaudina Molpadioide.Methods:Determined activity on total saponins according to 41.7%purity.Did the hemolytic test with "the tests of hemolysis and condensation" while acute toxicological experiment with <GB 15193.3 2003 acute toxicity tests>.Results:Partial hemolysis was found when eventual concentration was 0.2 mg/ml in 120 min while complete hemolysis in 180 min.With the maximum tolerance dose for 8000 mg/kg,the empty stomach of mice were irrigated through the mouth and nothing was found unnormal within 14 days of continuous observering.Conclusion:The total saponins from Acaudina Molpadioide holded its good ability to hemolysis,which may be related to the saponins structure.It probably because the saponins could caused the rabbit red cell membrane cracking happens,leaded to hemolysis.The saponins probably could been hydrolyzed which brought about changes of structure after the gastrointestinal tract of animal digestion and decomposition which implied that the total saponins was free from toxicity in a certain range.更多还原
