气相色谱质谱联用及电喷雾质谱法分析多棘海盘车的脑苷脂结构

【摘要】 目的 :鉴定多棘海盘车中脑苷脂的结构。方法 :通过反复硅胶柱层析法分离多棘海盘车中的脑苷脂,半制备高效液相色谱纯化。并采用气相色谱质谱联用(GC-MS)及电喷雾质谱(ESI-MS)的方法分析其结构。结果 :GC-MS分析表明,两种多棘海盘车脑苷脂MC-1、MC-2的糖基组成相同,均为葡萄糖;主要的长链碱均为d18:2和d18:3,还有少量的d22:1;两者结构最主要的差别在脂肪酸部分,MC-1由C2位不含羟基的脂肪酸组成,而MC-2的脂肪酸为C2位含一个羟基取代的脂肪酸。通过ESI-MS测定MC-1和MC-2的分子量范围为740~850之间。结论 :通过GC-MS和ESI-MS,初步鉴定了两种脑苷脂的结构和分子组成,为其进一步构效关系的研究提供结构依据。
【Abstract】 Objective:To determine the structure of the cerebrosides in the starfish Asterias amurensis.Methods:The cerebrosides were isolated through a silica gel column and were further purified by semi-preparative HPLC.The purified cerebrosides were further analyzed by Gas Chromatography-Mass Spectrometry(GC-MS) and electrospray ionization mass spectrometry (ESI-MS).Results:The GC-MS rusults suggested that the LCBs and glycoside composition of MC-1 and MC-2 are similar but with a difference in their fatty acid composition.The glycoside is glucose and the major LCBs are dl8:2 and dl8:3 with a few amount of d22:l.Compared to MC-1, the fatty acids of MC-2 were hydroxylated in the C2 position.The ESI-MS spectra shows the molecular weights of MC-1 and MC-2 are arised from 740-850.Conclusion:The basic structure of the cerebrosids can be confirmed by the methodology,and the experiment provided the basis for structure-activity foundation.
