4种水产动物源细菌主要分泌性蛋白的鉴定及其分泌性序列的分析

【摘要】 从本实验室分离保存的3种水产动物病原菌鳗弧菌(V.anguillarum)MN、鳗弧菌(V.anguillarum)3101、费氏弧菌(V.fischeri)及对虾肠道中分离的1种非病原菌坚强芽孢杆菌(Bacillus firmus)培养液中分别获取了分泌性蛋白,进行SDS-PAGE,并将表达量高的蛋白进行MALDI-TOF/TOF质谱鉴定。结果显示,有6条主要蛋白带被鉴定出来,分别是鳗弧菌金属蛋白酶、鳗弧菌锌金属蛋白酶、坚强芽孢杆菌甲壳素酶、坚强芽孢杆菌肠毒素A、费氏弧菌VFMJ111094蛋白和费氏弧菌ES114的外膜蛋白。将鉴定出的蛋白序列分别与NCBI数据库中的序列进行比对,并应用在线软件SignaIP 3.0分析信号肽序列,结果共筛选出了6条分泌性序列。用在线软件PSORT对分泌序列在细胞中的定位进行了分析,为高表达分泌性载体的构建找到了理论依据。
【Abstract】 The secreted proteins of Vibrio anguillarum MN,V.anguillarum 3101,V.harveyi and Bacillus firmus which were isolated and conserved by our laboratory,were extracted.The components of secreted proteins were separated by SDS-PAGE.The 6 high expressed protein bands in the SDS-PAGE were identified by mass spectrometry MALDI-TOF/TOF as the metalloprotease of Vibrio anguillarum,the zinc metalloproteinase of Vibrio anguillarum,the putative chitinase and the enterotoxin A of Bacillus firmus,the hypothetical protein VFMJ111094 and outer membrane protein of V.fischeri ESI 14.After downloaded the sequences of above proteins from NCBI database,6 signal peptides were given by using of the online software SignalP 3.0.The cellular localization of the secreted sequences were analyzed by PSORT.The results may be useful for construction of a secreted vector.
