牡蛎养殖过程中副溶血弧菌流行情况调查

【摘要】 为了研究牡蛎(Oyster)养殖过程中牡蛎中副溶血弧菌弧菌(Vibrio parahaemolyticus)含量的变化和副溶血弧菌含量与水质因子间的关系。用荧光定量PCR方法,对于2009年3月到2009年11月从广东阳西某牡蛎养殖场共采集到的26批,共178份牡蛎样品中副溶血弧菌的污染情况进行监测,比较了不同月份间,牡蛎中副溶血弧菌含量的变化;同时对温度、盐度、pH和溶解氧与副溶血弧菌含量之间的关系进行了分析。其中168份检测到了副溶血弧菌,检出率为94.38%;其中52份检测到了致病性副溶血弧菌,检出率为29.77%。牡蛎中总的副溶血弧菌含量从3月到8月呈上升趋势,随后呈下降趋势。牡蛎中致病性副溶血弧菌主要在6、7月被检出。对水温、盐度、pH和溶解氧与副溶血弧菌浓度进行相关性分析,结果表明,水温和副溶血弧菌浓度的相关性极显著(P<0.01),盐度和副溶血弧菌浓度的相关性显著(P<0.05),pH和溶解氧与副溶血弧菌浓度的相关性不显著(P>0.05)。对水温、盐度、pH和溶解氧与副溶血弧菌浓度进行回归分析,水温(T)和副溶血弧菌浓度(lgVP)的拟合回归方程为:lgVP=0.093×T-0.685(P=0.002,R~2=0.336);盐度(S)和副溶血弧菌浓度(lgVP)的拟合回归方程为:lgVP=1.199+0.116×S-0.004×S~2(P=0.05,R~2=0.389);水温(T)和盐度(S)对副溶血弧菌浓度(lgVP)的拟合回归方程为:lgVP=-1.941+0.116×T+0.058×S-0.001×S~2(R~2=0.414)。

【Abstract】 In order to study the changes of Vibrio parahaemolyticus during the oysters cultivation and the relationships between Vibrio parahaemolyticus and water factors.We collected 178 oyster samples in Yangjiang(Guangdong) from March 2009 to November 2009.We detected Vibrio parahaemolyticus of these samples by fluorescent quantitative PCR.We compared the different contents of Vibrio parahaemolyticus between every month.Also,we analyze the relationship between water factors(temperature,salinity,pH and dissolved oxygen) and Vibrio parahaemolyticus.In this study,168 of 178 samples were postive when they were tested by real-time PCR,the detection rate was 94.38%;There were 52 oyster samples contained pathogenicity VP,the accurate detection rate was 29.77%.Total Vibrio parahaemolyticus amount in Oyster increased from March to August,declined afterwards.The pathogenic Vibrio parahaemolyticus was detected in June and July.The relationship between the concentration of Vibrio parahaemolyticus and water temperature,salinity,pH and dissolved oxygen were analysed.The results showed that the water temperature and salinity with the concentration of Vibrio parahaemolyticus were obvious(P<0.01),pH and dissolved oxygen with the concentration of Vibrio parahaemolyticus were no significant(P> 0.01).We made a regression analysis between water temperature,salinity and pH and dissolved oxygen concentration with Vibrio parahaemolyticus.The regression equation of water temperature and the concentration of Vibrio parahaemolyticus is:lgVP = 0.093×T-0.685(P = 0.002,R2 = 0.336);The regression equation of Salinity and the concentration is:lgVP = 1.199 +0.116×S-0.004×S2(P = 0.05,R2 = 0.389);
