碳酸盐碱度对钝顶螺旋藻生长的影响

【摘要】 研究了不同碳酸盐碱度浓度条件下,钝顶螺旋藻OD560值、藻液pH值及碳酸盐碱度的变化。结果显示:培养4d后,4mg.·L-1和8mg·L-1组生长缓慢且呈现下降趋势,OD560值均低于对照组,具有极显著差异(P<0.01),16mg·L-1和32mg·L-1组与对照组相比,无显著差异,而64mg·L-1组具有显著差异(P<0.05)。第6~12d,4 mg·L-1、8mg·L-1组和16mg·L-1OD560值均低于对照组,具有极显著差异性(P<0.01),32mg·L-1组无显著差异,而64mg·L-1组在6~8d,具有显著差异(P<0.05),且在第10~12d,与其它实验组和对照组相比,表现出极显著差异性(P<0.01)。NaHCO3与Na2CO3组成的缓冲体系,其pH值调节功能优于单一使用NaHCO3的对照组。碳酸盐碱度测定结果表明,在充气状态下,实验期间各组的碱度始终保持动态平衡。

【Abstract】 Spirulina platensis was cultivated,in comparative studies,using different concentrations of carbonate alkalinity.The influence of carbon concentrations of NaHCO3+Na2CO3 on the growth of Spirulina platensis were studied in this paper.There were five treatment groups and one control set in this experiment. The treatment groups were 4mg·L-1,8mg·L-1,16mg·L-1,32mg·L-1,64mg·L-1 in carbonate alkalinity, respectively.NaHCO3 and Na2CO3 were added into the treatment group’s medium,while single NaHCO3 was added into the control medium based on the modified CFTRI medium which is widely used as the culture medium for Spirulina platensis.There were three parallels for both treatment groups and the control. The results showed that there were absolutely significant differences in the OD560 values between treatment groups of 4mg·L-1,8mg·L-1 and the control(P<0.01) on fourth day.The OD560 values in these two groups were lower than that of the control,while there were no significant differences between treatment groups of 16mg·L-1,32mg·L-1 and the control(P>0.05 ).There were significant differences between 64mg·L-1 and the control(P<0.05).The OD560 values in 4mg·L-1,8mg·L-1,16mg·L-1 groups were lower than that of the control from the sixth day to the twelfth day with significant differences(P<0.01).There was no significant difference between treatment group of 32mg·L-1 and the control,while significant difference was observed between 64mg·L-1 and the control from the sixth day to the eighth day(P<0.05).Moreover,there were absolutely significant differences(P<0.01 )in the OD560 values between the treatment group of 64mg·L-1 and other treatment groups and the control from the tenth day to the twelfth day.The OD560 values in 64mg·L-1 group were markedly higher than those of other treatment groups and the control from the tenth day to the twelfth day.It was pointed out that when compounds of NaHCO3 and Na2CO3 were added into the treatment group’s medium,the buffer functions for pH value were preferable to that of the control medium (modified CFTRI medium) in which single NaHCO3 was added.The pH value in treatment groups were kept balance at 8.59.8,while the pH value in the control kept a high value,which is reach over 10.23.The results also indicated that the carbonate alkalinity of HC03-+ CO32- were maintained balance when CO2 was pumped into the medium.It showed that high level CO2 in medium was transferred to carbonate resource and to HCO3- + CO32+.
