斜带石斑鱼雌激素受体的克隆、序列分析及表达模式的研究
Molecular cloning,characterization and expression profiles of three estrogen receptors in protogynous hermaphroditic orange-spotted grouper {Epinephelus coioides)

【摘要】 雌激素通过雌激素受体的介导,在脊椎动物的生殖系统中发挥着十分重要的生理功能。本研究利用RACE技术从先雌后雄雌雄同体的斜带石斑鱼(Epinephelus coioides)性腺中克隆到了三种雌激素受体的cDNA全长序列(gERα,gERβ1,and gERβ2),序列全长分别为2235bp,1967bp和21 58bp。分析显示这三种雌激素受体与相应的亚型相聚类。氨基酸序列比对分析表明获得的雌激素受体序列都具有其他物种雌激素受体的保守功能区。利用RT-PCR检测这三种雌激素受体在各种组织中的表达情况,结果表明这这三种雌激素受体在所检测的组织中都有表达,但表达水平具有明显差异。同时利用荧光定量PCR检测了这三种雌激素受体在卵巢发育和人工性逆转过程中的表达模式。结果显示三种雌激素受体显现出三种不同的表达模式,gERα在卵子成熟的过程中高表达,但在性逆转过程中急剧下降;gERβ1主要是在卵巢发育的早期表达较高,并且在性逆转过程中持续升高;gERβ2的表达变化波动较大。根据表达模式,推断出这三种雌激素受体在卵巢发育和性逆转过程中发挥着不同的生理功能。
【Abstract】 Estrogen plays key roles in vertebrate reproductive system via estrogen receptors(ERs) mediated pathway.In the present study,three full-length ERs cDNAs were isolated from the protogynous hermaphroditic orange-spotted grouper(Epinephelus coioides) and designated as gERa,gERpi,and gERβ2. The full length of gERs cDNA sequences were 2235bp for gERα,1967bp for gERβ1 and 2158bp for gERβ2, respectively.Phylogenetic analysis showed that each gER are clustered within the corresponding groups.By amino acid alignment analysis,gERs exhibited high evolutional conservation in functional domains with ERs in other species.Tissue expression studies by RT-PCR revealed that gERs expressed in all the obtained tissues.The expression profiles of three gERs in different ovarian stages by quantitative real-time RT-PCR. gERαwas highly expressed in the ovarian mature stages,and gERβ1 mainly expressed in the early gonad stages.The expression of gERβ2 was varying greatly during the ovarian development.Furthermore,the expression profiles of gERs during female to male sex reversal induced by 17α-methyltestosterone(MT) were also studied.The expression of gERαgradually decreased during the female to male sex reversal,while gERβ1 increased gradually.The expression of gERβ2 firstly decreased in female stage and recovered in male stage.
