两种罗非鱼GHSR基因5’侧翼区域的克隆及序列分析

【摘要】 生长激素促分泌素(ghrelin)是近几年才发现的生长激素促分泌素受体(GHSR)的内源性配体,ghrelin分布广泛,而且具有除促进生长激素分泌外的多种生理功能。前人的研究已经证明了ghrelin/GHSR系统的重要功能,但对于ghrelin与GHSR的转录调控却了解较少。本实验利用基因组步移法获得了两种罗非鱼GHSR基因的5’侧翼序列。莫桑比克罗非鱼(Oreochromismossambicus)GHSR基因的5’侧翼区域序列长1627bp,荷那龙罗非鱼(Oreochromis hornornum)的长度为1616bp。序列分析表明:两种罗非鱼GHSR基因5’转录调控区与黑鲷、人的GHSR基因5’侧翼区域有很多相同的转录调控元件,且均不含TATA盒;同时两种罗非鱼的5’侧翼区域具有自身特有的转录调控元件。
【Abstract】 Ghrelin,the recently discovered endogenous ligand for growth hormone secretagogue receptor (GHSR),is widely expressed and involved in regulating diverse physiological functions in addition to stimulation of growth hormone(GH) secretion.Previous studies have demonstrated the functional significance of the ghrelin/GHSR system.Using genomic walking,the 5’flanking region of GHSR was isolated from the genome of mozambique tilapia and Zanzibar tilapia.The 5’flanking region of mozarnbique tilapia GHSR is 1627bp in length,the Zanzibar tilapia’s is 1616bp in length.Sequence analysis indicated that the two tilapia,seabream and human GHSR 5’flanking region shared several conserved binding sites for transcription factors,and they all have no TATA box;the tilapia GHSR has unique putative transcription factor-binding sites different from the human and seabream counterpart.
