基于形态特征和线粒体cytb基因序列探讨棱鳀属的系统进化

【摘要】 对6种棱鳀鱼类共13个个体的cytb基因进行PCR扩增,经比对校正得到1143bp的基因片段。序列变异,碱基含量,Kimura双参数法遗传距离和NJ树均有MEGA3.1软件构件。在13个序列中共检测到361个变异位点,序列中转换多于颠换,平均碱基含量分别为27.8%T,29.5%C,26.7%A和15.9%G。A+T含量(54.5%)明显高于C+G含量(45.5%)。基于Kimura双参数法,计算的种间遗传距离从0.5%(黄吻棱鳀与中颌棱鳀)到21.7%(黄吻棱鳀与赤鼻棱鳀)。黄吻棱鳀和中颌棱鳀间的遗传距离近似或者等于种内的遗传距离(0.3%~0.5%)。以日本鳀为外群构建NJ树。进化树显示,黄吻棱鳀和中颌棱鳀关系最近。通过形态和线粒体cytb基因片断分析,本研究分子数据结果支持中颌棱鳀和黄吻棱鳀应为为同一个种,但是对于汉氏棱鳀和杜氏棱鳀的分类地位尚待进一步的解决。

【Abstract】 A molecular phylogenetic analysis was undertaken to evaluate the phylogenetic relationship of 6 species of Thryssa in China,the mitochondrial cytb genes of 13 individuals were amplified using PCR techniques.About 1300bp length fragments were obtained and sequenced.After aligned,a fragment of 1143 bp nucleotide sequence which could be used for sequence analysis was obtained.The genetic information indexes,including Kimura-2 parameter genetic distance,basal composition were generated using MEGA3.1 software and phylogenetic tree was reconstructed by tree finder software.Of the 1143 aligned base pairs,361 sites were variable and the mutation sites were mainly transition and transversion.Transitions were found to be more common than transversions which suggested that the sequences had not reached saturation.No insertions/deletions were detected.The average basic compositions of all sequences were as follows 27.8%T, 29.5%C,26.7%A,and 15.9%G.The genetic distances between the six species ranged from 0.5%(Thryssa mystax and Thryssa vitrirostris) to 21.7%.Molecular phylogenetic tree was constructed by NJ methods based on the 1146 bp cytb sequence,with Engraulis japonius as the designated outgroup.The most noteworthy findings were that the genetic distance between two species(Thryssa mystax and Thryssa vitrirostris),which had identical morphological characteristics,was identical with that within species. Moreover,the NJ tree indicated a closer relationship between Thryssa mystax and Thryssa vitrirostris.So we suggest that the Thryssa mystax and Thryssa vitrirostris might be one species.
