大口黑鲈选育群体和养殖群体的遗传多样性研究分析

【摘要】 本研究采用AFLP技术对加州鲈鱼非选育群体和第三,四代选育群体的遗传多样性及遗传分化进行分析,计算了3个群体间的遗传多态度、遗传距离及分化系数。结果显示,8对引物共产生了262条带,其中多态性条带有80条,平均每对引物检测到10条多态性条带。3个群体的多态性位点比例分别为30.29%、29.36%、29.20%;Shannon多样性指数分别为0.2042、0.2017、0.1955.群体间的遗传变异(HSP-HPOP)/HSP平均为0.0752,由此可见,92.5%的遗传变异来自于群体内,而7.5%的遗传变异来自于群体间。第三,第四代选育群体和非选育群体相比遗传多样性略有下降,并显示了加州鲈鱼选育群体还有较大的选育潜力,可以继续保持遗传响应,最终保证选育工作的成功。

【Abstract】 The AFLP technique was used to analyze the genetic diversity and genetic differentiation of a unselected population and 2 consecutive selected populations of largemouth bass(Micropterus salmoides).The genetic polymorphisms,genetic distances and differentiation indexes were calculated.These results showed that 262 bands were generated with 8 primer sets,and 80 bands were polymorphic bands.On the average,every primer set generated 10 polymorphic bands.The polyrnorphic loci of the three populations accounted for 30.29%,29.36%and 29.20%in all detected loci.The genetic diversities of the unselected population and the selected populations quantified by Shannon indexs were 0.2042,0.2017 and 0.1955,respectively.The inter-population and intra-population variance of genetic diversity was 0.0752 and 0.9248,respectively...Compared with cultivated population,the selected population of the third and fourth generation showed a slight decrease of genetic diversity.However the two selected populations had great potential for selective breeding of the largemouth bass(Micropterus salmoides.)
