剑尾鱼分泌型IgM在初次和再次免疫中的动态变化

【摘要】 目的:剑尾鱼(Xiphophorus helleri)腹腔注射免疫嗜水气单胞菌(Aeromonas hydrophila)灭活疫苗后,应用荧光定量PCR检测分泌型免疫球蛋白M(sIgM)mRNA在脾脏和头肾中的变化情况,探讨剑尾鱼作为疫苗评价模型的可行性。方法:剑尾鱼注射嗜水气单胞菌灭活疫苗后分别在第一次免疫后1、4、7d,加强免疫后的4、7、1 1d,取脾脏和头肾两种组织,用实时荧光定量PCR方法检测其sIgMmRNA的转录水平。结果:脾脏和头肾中显示相似的动态变化,均在第一次免疫后4d达到高峰,之后在加强免疫后4d sIgM mRNA的表达量又出现一个高峰,然后逐渐下降。结论:剑尾鱼在第一次免疫、第二次免疫后脾脏、头肾组织中均检测到了IgM表达上调表明,分泌型免疫球蛋白(sIgM)可能既参与先天性免疫,又参与获得性免疫,为剑尾鱼作为鱼类疫苗评价模型的研究奠定了一定的基础。

【Abstract】 Objective:to study the transcription changes of sIgM mRNA in spleen and head kidney of immunized swordtail(Xiphophorus helleri) with q-PCR after following injection procedures applying preparations of formalin- killed bacteria(Aeromonas hydrophila).Study on the feasibility of swordtail (Xiphophorus helleri) being vaccine evaluation model.Method:Transcription of mRNA was measured in spleen,head-kidney samples taken 1,4,7d post- injection and taken 4,7,lld in second-injection using q-PCR.Result:Spleen and head kidney showed similar changes,in which a peak presented at the 4th day post- injection and the second peak at the 4th day in second-injection,then the expression of sIgM mRNA gradually decreased.Conclusion:These results indicated the up-regulation of swordtail sIgM expression in spleen and kidney by bacterial vaccine occurred at the phase of the primary immunity and the secondary immunity,suggesting that sIgM in swordtail may have a role in innate immunity and adaptive immunity.It contributes for swordtail(Xiphophorus helleri) being fish vaccine evaluation model.
