【摘要】 目的通过体外酶学试验研究白艾提取液对黄嘌呤氧化酶活性的影响及其酶抑制机制。方法运用紫外分光光度计观测尿酸生成的吸光度变化值,测定波长为292nm,并以黄嘌呤为底物绘制Lineweaver-Burk和Dixon图。结果白艾提取液对黄嘌呤氧化酶具有较强的抑制作用,其Ki值为11.38μg/ml,抑制类型为混合型。而别嘌呤醇作为参照,其Ki值为0.62μg/ml,抑制类型为竞争性抑制。结论白艾提取液治疗痛风的作用机理之一是抑制黄嘌呤氧化酶活性。更多还原

【Abstract】 To assay the xanthine oxidase(XO) inhibitory activity of the Gnaphalium affine extract in vitro and to determine its enzyme inhibition mechanism.Xanthine oxidase inhibitory activity was assayed spectrophotometrically under aerobic conditions and the degree of enzyme inhibition was determined by measuring the increase in absorbance at 292 nm associated with uric acid formation.Enzyme kinetics was carried out using Lineweaver-Burk and Dixon plots using xanthine as the substrate.The Gnaphalium affine extract exhibited strong effect on XO with a Ki value of 11.38μg/ml.In comparison,the known synthetic XO inhibitor allopurinol, used as a reference standard,showed a Ki of 0.62μg/ml.Enzyme inhibition mechanism indicated that the mode of inhibition was found a Mixed type.These results suggest that the use of Gnaphalium affine for the treatment of gout could be attributed to its xanthine oxidase inhibitory activity.更多还原
