结核病患者血清蛋白质/多肽谱的初步研究并文献复习

【摘要】 目的了解结核病患者血清蛋白质/多肽的表达,为新型结核病血清学诊断方法的研究提供基础依据。方法 (1)应用WCX蛋白质芯片和表面增强激光解析-电离飞行时间质谱(SELDI-TOF-MS)技术对99例肺结核患者和18例肺外结核患者进行了血清蛋白质/多肽谱检测,采用ClinProTools分析软件进行比较分析。(2)以"结核"、"双向电泳"、"表面增强激光解析-电离飞行时间质谱"、"血清学"、"蛋白质组学"及相应英文为检索词在万方医学数据库、PUBMED中检索中、外文献进行文献复习。结果 (1)WCX蛋白质芯片从血清中捕获到99种蛋白质/多肽,其中m/z为5753.76的蛋白质在肺结核与肺外结核两组间存在显著性差异,在肺外结核患者血清中表达上调,其平均质谱峰面积在肺结核患者和肺外结核患者中分别为15.42±4.86、28.29±14.08。(2)检索到密切相关文献4篇(其中英文文献1篇、中文文献3篇),均提示肺结核患者与健康对照者间血清蛋白质组存在差异。结论结核病的发生可产生大致相同的血清蛋白质/多肽谱,在不同类型结核结核病患者血清中可以找到共同的血清学诊断标志物。

【Abstract】 Objective To understand the expression of serum protein/ peptide of the patients with pulmonary tuberculosis so as to provide a basis for serological diagnostic method of new style tuberculosis.Medthods(1) The serum protein/ peptide mapping of 99 patients with pulmonary tuberculosis and 18 patients with extrapulmonary tuberculosis were detected by using the techniques of WCX protein chip and surface-enhanced laser desorption and ionization time of flight mass spectromerty(SELDI-TOF-MS ).The powervision of ClinProTools was employed to give comparative analysis.(2) The terms,containing tuberculosis,two-dimensional electrophoresis,SELDI-TOF-MS,serology,proteomics and other related terms were searched in Wanfang medical data base and PUBMED,by which a few Chinese literatures and foreign languages literatures were reviewed.Results(1) 99 proteins/ peptides in serum were captured by the technique of WCX protein chip, among which there was a significant difference in the protein of m/z(5753.76) between the pulmonary tuberculosis group and the extrapulmonary tuberculosis group.The expression of m/z(5753.76) in serum of the patients with extrapulmonary tuberculosis was been upregulation and the average peak areas of mass spectrum (MS) in the pulmonary tuberculosis group and the extrapulmonary tuberculosis group were 15.42±4.86, 28.29±14.08,respectively.(2) 4 related literatures(1 English literature and 3 Chinese literatures) were found, suggesting that there were significant differences in the serum proteome between the patients with pulmonary tuberculosis and the healthy controls.Conclusion The incidence of tuberculosis may produce roughly the same serum protein / peptide mapping and the common diagnostic markers of serology can be found in the serum of different types of tuberculosis patients.
