口腔黏膜体外三维培养模型的建立

【摘要】 目的:建立口腔黏膜体外三维培养模型。方法:体外分别培养牙龈上皮细胞及成纤维细胞备用。胶原溶液与成纤维细胞按比例混合后置于Transwell小室培养两天后,于胶原-细胞混合体表面接种上皮细胞,共同增殖培养三天后,升至液气平面继续培养十天。组织学处理,染色检测三维模型的组织学结构。结果:HE检测可见口腔黏膜三维培养模型具有良好分化的上皮结构及固有层成分,结构类似天然口腔黏膜结构。结论:利用Transwell小室及胶原基质,采用增殖-分化培养方法,可得到类似天然口腔黏膜结构的三维培养物。
【Abstract】 Objective:To establish a three-dimensional oral mucosa model in vitro.Method:Culture the gingival epithelial cells and fibroblasts in vitro respectively.Mix the collagen solution and fibroblasts,and then seed the mixture into the Transwell.After two day’s of culture,seed the epithelial cells onto the surface of the dermal equivalent(collagen-fibroblast mixture).Proliferation culture for three days,then lift the model to an air-liquid surface for another 10 days.Harvest the composite culture,and then determine the historical characteristics of the three-dimensional model by staining.Results:HE staining shows that the composite culture has well-differentiated epithelial and lamina propria structure,which is quite similar to the natural oral mucosa.Conclusion:By application of Transwell,collagen solution and the proliferation-differentiation culture method,we can get a stable and satisfactory three-dimensional oral mucosa model.
