MMP9敲除导致实验诱导根尖周炎病损面积增大

【摘要】 目的:比较MMP9敲除小鼠相对于野生型小鼠根尖周炎病损进展过程的区别。方法:对小鼠下颌第一磨牙进行牙髓开放,分别在术后0,7,14,21,28天后处死动物并收集标本。HE染色观察牙髓,根尖,根尖周特征以及根尖周缺损面积。显微CT进一步观察计量根尖周缺损面积。抗酒石酸染色观察破骨细胞的数目。荧光定量PCR和免疫组化分别检测RANK,RANKL,OPG,IL-1β,TNF-α,MMP2和MMP8在RNA水平和蛋白水平的表达量。结果:MMP9敲除小鼠的根尖周缺损面积明显大于野生型小鼠(P<0.05)。两种小鼠的牙髓、根尖、根尖周特征以及根尖周缺损区破骨细胞的数量没有显著差异(P>0.05)。MMP9敲除小鼠根尖周病损区中细粒细胞的数量高于野生型小鼠(P<0.05),其RANK,RANKL,OPG,IL-1β,TNF-α,MMP2以及MMP8的表达水平也高于野生型小鼠(P<0.05)。结论:MMP9敲除小鼠在实验诱导形成的根尖周炎中根尖周病损区面积比野生型小鼠大,并且伴发更为严重的炎症反应。这意味着MMP9在宿主对抗根管内以及根尖周组织感染的免疫和炎症反应中发挥重要作用。

【Abstract】 Objective:To evaluate the lesion progression in MMP9 knockout(MMP9 KO) mice compared with that in control mice(WT).Methods:The pulps of mouse mandibular first molars were exposed;animals were killed at 0,7,14,21,and 28 days after surgery.Hematoxylin-eosin-stained sections were observed for the description of pulpal,apical,periapical features,and the periapical lesion size.The periapical lesion size was further measured with micro-CT.The number of osteoclasts was also counted by tartrate resistant acid phosphatase histoenzymology.Real-time Polymerase Chain-reaction and Immunohistochemistry were used to analyze the expression level of RANK,RANKL,OPG,IL-1β,TNF-α,MMP2 and MMP8.Results:There was a significant difference(P<0.05) between the 2 types of animals regarding the periapical lesion size,which was larger in MMP9 KO animals.No significant differences(P>0.05) were found between WT and MMP9 KO mice related to the osteoclast number as well as the pulpal,apical,and periapical features.More neutrophil cells were seen in MMP9 KO animals than WT mice(P<0.05).The expression levels of RANK,RANKL,OPG,IL-1β,TNF-α,MMP2 and MMP8 were found up-regulated in MMP9 KO mice(P<0.05).Conclusions:MMP9 KO animals developed larger periapical lesions with greater inflammatory response,indicating an important role of MMP9 in the host’s immune and inflammatory response to root canal and periradicular infection.
