载药等离子体对粪肠球菌生物膜的作用

【摘要】 目的:评估大气压非平衡态载药等离子体对体外形成的粪肠球菌生物膜的灭菌效果。方法:无菌盖玻片置于1 2孔板中,将粪肠球菌悬液接种于盖玻片上,厌氧环境下37℃培养1周。形成粪肠球菌生物膜后,进行如下处理:大气压非平衡单纯等离子体或载药(洗必泰葡萄糖酸盐)等离子体处理1或5分钟,以0.2%洗必泰葡萄糖酸盐溶液漂洗和生理盐水漂洗作为对照;应用荧光染料对处理后的生物膜进行染色,激光扫描共聚焦显微镜下观察灭菌效果并作统计分析。结果:对于相同处理时间来说,载药等离子体组的灭菌效果明显优于单纯等离子体组和洗必泰漂洗组(p<0.05),与洗必泰漂洗和载药等离子体共同处理组的灭菌效果相同(p>0.05)。单纯等离子体组、载药等离子体组和洗必泰冲洗组的灭菌效果随暴露时间的延长而增加(p<0.05)。结论:大气压非平衡态载药等离子体具有良好的体外灭活粪肠球菌生物膜的作用,可以作为根管治疗过程中一个有效的辅助治疗手段。

【Abstract】 OBJECTIVE:The aim of this study was to evaluate the in vitro antimicrobial effect by atmospheric pressure non-equilibrium plasmas(APNPs)with chlorhexidine digluconate(CHX)against Enterococcus feacalis biofilm.METHODS:Sterile coverslips were placed in the wells of tissue culture plate exposing to Enterococcus faecalis for 1 week at 37℃to form a biofilm.The bacterial biofilms were processed to APNPs(8 kV,8 kHz,1600 ns,He/O2:1:0.01 L/min)with or without 5%CHX for 1 and 5 minutes,whilst subjected to 0.2%CHX or saline irrigating for corresponding times as control.Following treatment,the death ratios indicating the dead bacteria to all of bacteria in biofilms were analyzed by viability staining and confocal laser scanning microscope(CLSM).RESULTS: For the same exposing time,the combined use of APNPs with 5%CHX had a more remarkable(p<0.05)antibacterial effect against the biofilms than either APNPs or 0.2% CHX irrigating alone,but the same(p>0.05)as the APNPs with 5%CHX after CHX washing.In addition,there was a more pronounced(p<0.05)antimicrobial effectiveness against bacterial biofilms treated by APNPs with or without CHX and CHX washing for longer exposing time.CONLUSION:The use of APNPs with chlorhexidine digluconate had a promoted antibacterial in vitro activity against Enterococcus faecalis biofilms,and could be applied to as a novel effective assistant tool against bacterial biofilms in course of root canal therapies.
