黏性放线菌与伴放线放线杆菌拮抗关系的体外研究

【摘要】 目的:研究黏性放线菌(A.v)与伴放线放线杆菌(A.a)体外培养的拮抗关系,在有益菌的替代疗法方面对治疗龋病和侵袭性牙周炎提供实验依据。方法:1.将A.v、A.a置于相同环境下培养,观察两细菌以及混合后的生长情况并绘制生长曲线,计数A.a在混合培养后占菌落总数的百分比。2.Sabi ne法将A.v不同浓度的全菌细胞菌悬液、粉碎液和上清液,滴加在已涂布A.a血琼脂板上培养,观察并测量其抑菌圈。同样的方法观察并测试A.a对A.v的抑制情况。结果:1.A.v生长速度较快,而A.a生长相对较慢。混合组的生长曲线与单菌种A.v较相似,而A.a呈现出不同的生长曲线。随着混合细菌组细菌培养的时间越长,A.a在血琼脂平板上所占菌落总数的百分比越小,呈下降趋势,差异具有统计学意义(P<0.05)。2.A.v的全菌细胞菌悬液对A.a有抑菌作用,抑菌圈平均直径(mm)为7.87、14.33、1 5.74和18.42,差异有统计学意义(P<0.05);A.v的粉碎液、上清液对A.a无抑制作用。A.a对A.v的抑菌实验中未见有抑菌圈的出现。结论:A.v对A.a的生长有抑制作用,提示A.v作为A.a的拮抗菌,用以限制侵袭性牙周炎的生态防治方面值得深入研究。

【Abstract】 OBJECTIVE:To study the antagonistic relationship between Actinomycetes viscous and Actinobacillus actinomycetemcomitans via in vitro experiment.The objectives were to provide experimental evidence for the treatment to dental caries and aggressive periodontitis by replacement therapy with beneficial bacteria.METHODS:1.The two strains were cultured under the same environment,the growth of the Actinomyces viscosus,Actinobacillus actinomycetemcomitans and two mixed were observed,and the growth curve was made,the shared percentage of Actinobacillus actinomycetemcomitans strains in total plate count after the two mixed cultured was counted.2.The Sabine method was implemented in this research.The bacterial suspension with whole bacterial cells with different concentrations,grinding fluid and supernatant of Actinomycetes viscous,were dropped on the blood agar plate that has been coated by Actinobacillus actinomycetemcomitans,anaerobic incubated,to observe and measure the inhibition zone. The same method was also used to observe and measur inhibition zon of Actinobacillus actinomycetemcomitans to Actinomyces viscosus.RESULTS:1.It is indicated that Actinomyces viscosus grew faster,whereas the Actinobacillus actinomycetemcomitans. growth was relatively slow.Growth curve of mixed group were similar with the single strain Actinomyces viscosus,and Actinobacillus actinomycetemcomitans showed different growth curves with the other two groups.lt is found that the longer incubation time,is in the less percentage sharing total colonies of Actinobacillus actinomycetemcomitans of mixed bacterial group on the blood agar plates,which showed a downward trend, the difference was statistically significant(P<0.05 ).2.The suspensions with whole-bacterial cells of Actinomyces viscosus has the inhibite effect on coated Actinobacillus actinomycetemcomitans,inhibition zone Average diameter(mm) for the 7.87,14.33,15.74 and 18.42,the difference statistically significant(P <0.05);grinding fluid and supernatant of Actinomyces viscosus no inhibition on Actinobacillus actinomycetemcomitans.Inhibition zone has not seen in inhibition experiments of Actinobacillus actinomycetemcomitans to Actinomyces viscosus.CONCLUSION:Actinomyces viscosus inhibit the growth of Actinobacillus actinomycetemcomitans.The new strategy of inhibiting Actinobacillus actinomycetemcomitans with Actinomyces viscosus as beneficial bacteria or aggressive periodontal replacement was worthy of further study.
