蜂房主要有效成分槲皮素对口腔致龋菌的影响

【摘要】 目的:蜂房提取物可以抑制变异链球菌等多种龋病相关菌的生长、产酸、生物膜形成。通过高效液相色谱和质谱分析发现,蜂房防龋有效组分中主要化合物为槲皮素。槲皮素是一种天然黄酮类化合物,对多种革兰阳性及阴性菌的生长均有抑制作用。本实验研究槲皮素对口腔常见致龋菌生长,产酸,耐酸的影响,及其与三氯生,西吡氯铵等口腔常用抗菌剂的协同作用。材料和方法:测定槲皮素对6种口腔内常见致龋菌的最低抑菌浓度(MIC)和最低杀菌浓度(MBC)以及亚抑菌浓度下药物对变异链球菌产酸,耐酸水平的影响。应用棋盘式微量液体稀释法及部分抑菌浓度指数(FICI)分析,测定其与三氯生和西吡氯铵的联合作用药效。结果:槲皮素对6种致龋细菌(变异链球菌,远缘链球菌,血链球菌,粘性放线菌,内氏放线菌,乳杆菌)的生长均有一定的抑制作用(MIC 1-4 mg/ml和MBC 4-1 6 mg/ml)。亚抑菌浓度的槲皮素可明显抑制变异链球菌的产酸性和耐酸性(6.74%-50.11%)。在抑制变异链球菌生长方面,槲皮素与西吡氯铵间存在协同效应(FICI<0.5)。结论:蜂房主要有效成分槲皮素能抑制变异链球菌等口腔常见致龋菌的活性及致龋毒力因子,并与西吡氯铵间存在协同效应。提示我们蜂房作为一种潜在的防龋药物可能有着良好的应用前景。
【Abstract】 Objective:NidusVespae(the honeycomb of Polistesolivaceous(De Geer), P.japonicus Saussure,and Parapolybiavariafabricius) as a kind of traditional Chinese medicine has been demonstrated to inhibit the growthand acid-production of oral cariogenic bacteria.We found the most abundant compound in NidusVespaeeffective fraction is quercetinby HPLC and Electrospray ionisation-mass spectrometry(ESI-MS) analysis.This study was toevaluate the effects of quercetin on oral cariogenic bacteria growth,acid production,acid tolerance activity and its synergy effect with triclosan and cetylpyridinium chloride(CPC).Materials and methods:Tested antimicrobial activity of quercetin(MICs and MBCs) against six cariogenic bacteria and its acid inhibition and acid tolerance properties against Streptococcus mutans.Further evaluated its synergy effect with triclosan and cetylpyridinium chloride(CPC) using fractional inhibitory concentration index(FIG) analysis.Results:Quercetininhibited the growth of oral cariogenic bacteria(S. mutans,S.sobrinus,S.sanguis,A.viscosus,A.naeslundii and L.rhamnosus) ranging from 1 to 4 mg/ml at minimum inhibitory concentrations and from 4 to 16 mg/ml at minimum bactericidal concentrations.Quercetinsignificantly inhibitedacidogenicity and acidurityof Streptococcus mutants at sub-MIC levels(6.74%-50.11%).There was synergy between quercetin and CPC against S.mutants(FICI<0.5).Conclusion:The results showed that quercetinconsist inNidusVespae presented remarkably biological activity,which suggests that NidusVespaemight be useful as a potent preventive and therapeutic agent in dental caries.
