抑制NF-κB传导通路对舌癌细胞凋亡的影响

【摘要】 目的:探讨抑制NF-κB/p65信号传导通路与平阳霉素诱导Tca8113细胞凋亡的关系。方法以脂质体为载体,转染2ug/ml的p65反义寡核苷酸于Tca8113细胞后,8ug/ml的平阳霉素处理细胞,免疫组化、Western blot法检测细胞核内p65含量变化,流式细胞术检测细胞凋亡率。结果:平阳霉素能激活Tca8113细胞内的NF-κB/p65信号传导通路,而p65反义寡核苷酸转染能明显抑制该信号通路,细胞凋亡率明显提高。结论:抑制NF-κB/p65信号传导通路能提高平阳霉素诱导的Tca8113细胞凋亡率。
【Abstract】 Objective:To examine the relationship between the inhibition of NF-κB signal transduction way and the apoptosis of Tca8113 Carcinoma Cell induced by Pingyangmycin chemotherapy Methods:2ug/ml antisense p65 oligodeoxynucle-otides(5′- GAACAGTTCGTCCATGGCCG-3′) was transfected into Tca8113 cells through Lepofectin vectin,then the cells were treated with 8ug/ ml Pingyangmycin.After 3h and 6h,the expression of p65 in nucleus was studied with immunohistochemical methods and Western blot analyses.After 48h,the apoptosis rate of cell was detected with flow cytometry analysis.Results:The NF-κB/p65 Signal Transduction way in Tca8113 cells was activated after treated with Pingyangmycin.But the transfection of antisense p65 oligodeoxynucle-otides inhibited the activation of the signal transduction,the p65 expression in nucleus was decreased significantly(11.95±0.97 vs 5.59±0.38×104) at 6h,and the apoptosis rate of cell was increased significantly(19.3±1.12%vs 42.23±3.47%) at 48h.Conclussion:The results suggested the apoptosis rate of Tca8113 carcinoma cell induced by Pingyangmycin was improved by the inhibition of NF-κB/p65 signal transduction.
