Id-1是治疗人涎腺腺样囊性癌的潜在新靶点

【摘要】 目的:腺样囊性癌(ACC)是一种复发率较高、虽然生长缓慢但是侵袭性很强的肿瘤。Id(inhibitor of DNA binding,DNA结合抑制因子/分化抑制因子)蛋白Id是一组对碱性螺旋-环-螺旋(basic Helix-Loop-Helix)活性起负调节作用的转录因子,可抑制细胞分化,促进细胞增殖。本实验拟研究Id-1在人涎腺腺样囊性癌中的基本功能。方法:应用Western blotting比较腺样囊性癌细胞系(ACCM)和其他三种细胞系(SAS,8113,ACC2)内Id-1蛋白的表达区别。然后,对ACCM行Id-1基因的RNA干扰;应用RT-PCR和Western blotting对干扰前和干扰后的Id-1进行RNA水平和蛋白水平的检测,以确认干扰效果。对干扰前和干扰后的ACCM分别行增殖能力、侵袭能力和凋亡水平的检测。结果:我们发现相比其他三种细胞系(SAS,8113,ACC2),在ACCM内Id-1蛋白是高表达的。RT-PCR和Western blotting结果显示对ACCM行Id-1基因的RNA干扰是有效的。RNA干扰后,ACCM的增殖能力和侵袭能力都大幅度下降;凋亡率大大提高。结论:在人涎腺腺样囊性癌的治疗中,Id-1是一个有潜力的治疗靶点。

【Abstract】 Purpose:Adenoid Cystic Carcinoma(ACC) is a slowly growing but highly invasive cancer with high recurrence rate.Id(Inhibitor of DNA binding) proteins are dominant regulators of basic helix-loop-helix transcription factors that control malignant cell behavior in many different tissues.This study aimed to identify the potential role of inhibiting DNA binding -1(Id-1) in human salivary adenoid cystic carcinoma(SACC) progression.Methods:First,we compared the Id-1 protein expression in adenoid cystic carcinoma cell line(ACCM)with other three cell lines(SAS,8113,ACC2)with Western blotting.Then we measured Id-1 mRNA and protein expression in ACCM before and after RNA interference(RNAi) with RT-PCR and western blotting.The proliferation and the ability of invasiveness of ACCM cells was measured through MTT and Boyden chamber invasion assays.We used flow cytometry to detect the apoptotic in ACCM before and after RNAi.Results:We found that the Id-1 protein expression in human salivary ACCM was extraordinary high.It showed successful knock down of Id-1 after RNAi.Further studies showed the proliferation and the ability of invasiveness of ACCM cells were dramatically down-regulated after RNAi.And more apoptotic cells were detected after Id-1 silencing.Conclusions: Our data suggest Id-1 is a potential target in the treatment of human salivary adenoid cystic carcinoma.
