RNA干扰iNOS基因抑制舌鳞癌细胞VEGF表达的实验研究

【摘要】 目的:探讨诱导型一氧化氮合酶(iNOS)基因对舌鳞癌细胞株Tca8113细胞中血管内皮生长因子(VEGF)的表达调控情况。方法:以RNA干扰技术将含有iNOS基因的特异性shRNA片断的质粒表达载体转染入Tca8113细胞中,RT-PCR和Western-blotting分别检测iNOS\VEGF的mRNA、蛋白表达改变情况。结果:Tca8113细胞的iNOS\VEGF基因的PCR产物,在转染后24h、48 h实验组和对照组之间比较,差异均有显著性(P<0.05);iNOS基因的蛋白产物,在转染后36 h、48 h差异均有显著性(P<0.05)。VEGF基因的蛋白产物,在转染后48 h差异有显著性(P<0.05)。结论:以RNA干扰技术沉默Tca8113细胞中iNOS基因,可以降低细胞中VEGF的表达水平,iNOS基因可能成为肿瘤防治的新靶点。

【Abstract】 Objective:To investigate the regulation of inducible nitric oxide synthase(iNOS )to vascular endothelial growth factor (VEGF) expression.Methods:we used RNA interference(RNAi) technique and transfected the vector which involves short hairpin RNA of iNOS to Tca8113 cell by LIPOFECTAMINE 2000,detected the iNOS and VEGF’s change in the gene and protein expression level by RT-PCR and Western-blotting for the sake of finding out if iNOS can regulate the expression of VEGF.Results:the VEGF expression in Psilencer-iNOS group Tca8113 cell decreased in both mRNA and protein level to a certain degree(P<0.05),besides the downexpression of iNOS by RNA interference to iNOS(P<0.05).Conclusion:There is relationship between iNOS and VEGF.iNOS gene RNAi can downexpression of VEGF in Tca8113 cell.
