涎腺粘液表皮样癌细胞诱导肿瘤血管内皮细胞三维成型的体外实验

【摘要】 目的:观察在三维胶原体外培养条件下涎腺粘液表皮样癌细胞体外诱导肿瘤血管内皮细胞血管生成的作用。方法:采用鼠尾胶原作为肿瘤血管内皮细胞培养的支架,以原代培养的粘液表皮样癌细胞作为诱导条件,观测应用免疫磁珠分选出的粘液表皮样癌血管内皮细胞的生长和小管样结构形成的过程,测定形成曲线、小管管径和管长以及细胞周期,比较有和无粘液表皮样癌细胞诱导肿瘤血管内皮细胞形成小管样结构的差异。结果:粘液表皮样癌细胞对体外三维培养肿瘤血管内皮细胞具有诱导小管形成的作用,所诱导的小管管径和管长数值均较对照组大,小管形成时间早。结论:粘液表皮样癌细胞能促进内皮细胞的生长、迁移和小管结构的形成,是研究粘液表皮样癌发展过程中促血管生成的体外良好模型。

【Abstract】 Objective:The study was to observe neovascularization with mucoepidermoid carcinoma vascular endothelial cells induced by mucoepidermoid carcinoma cells in the three-dimensional collagen matrix model in vitro.Methods:The collagen culturing for mucoepidermoid carcinoma vascular endothelial cells was made up of rat tail tendon collagen.With induction by cells of primary mucoepidermoid carcinoma,the current research observed growth of mucoepidermoid carcinoma vascular endothelial cells that were purified by immunomagnetic beads and formation of tubule-like structures(TLSs).It observed the difference of TLSs of vascular endothelial cells with or without induction of mucoepidermoid carcinoma cells in terms of the growth curve,the diameter and length of the TLSs, and the cell cycle.Results:The mucoepidermoid carcinoma cells could induce formation of TLSs in the three-dimensional collagen matrix model.The diameters and lengths of TLSs in the induction group were larger than those in the control group(p1<0.05,p2<0.05); and the TLSs were formed at the earlier time(p<0.05).Conclusion:The mucoepidermoid carcinoma cells could promote growth and migration of mucoepidermoid carcinoma microvascular endothelial cells,as well as the formation of TLSs.The three-dimensional collagen matrix model with induction of mucoepidermoid carcinoma vascular endothelial cells by mucoepidermoid carcinoma cells in the current research may be a favorable choice for research on pro-angiogenic factors for progression of mucoepidermoid carcinoma.
