静止层位置对于细胞电泳仪测量精度的影响
THE INFLUENCE OF THE STATIONARY LEVEL LOCATION ON THE MEASUREMENTAL ACCURACY OF A RED CELL ELECTROPHORESIS APPARATUS

【摘要】 文中介绍扁平形、长方形和圆形横截面的细胞电泳小室中静止层位置的计算方法,指出对于正方形横截面说来,由壁面起算的静止层深度与小室高度之比应为△≈1/8,某些产品中通用的△=1/4是错误的。现有装置中静止层附近介质电渗速度变化较快,因而显微镜聚焦深度定位不准对于细胞电泳速率测量结果影响较大。作者建议设计双管式电泳仪,可使静止层附近的电渗速度始终保持几乎为零,从而大大减小深度定位不准所造成的测量误差。
【Abstract】 This paper presents the methods for calculating the locations of the stationary levels in the red cell electrophoresis apparatuses with flat,rectangular or circular cross sections.It is indicated that,for a cross section of square shape.the correct value of the relative depth of the stationary level (scaled to the height)is△≈1/8 rather than the commonly misused△=1/4 in some commercial apparatuses. It is also shown that,in the available apparatuses,the inaccuracy of the microscopic focusing may lead to significant errors in the observed rates of the cell electrophoresis,due to the rapid change of the electroosmotic velocity at the neighborhood of the stationary level,The suggested design of a double tube electrophoresis apparatus is expected to produce a zone of almost vanishing electroosmotic velocity near the stationary level so that the above mentioned errors may be minimized.
