TSA提高肝癌细胞Be17402细胞对TRAIL的敏感性

【摘要】 目的观察TSA是否能提高肝癌细胞Be17402对肿瘤坏死因子凋亡相关配体(TRAIL)的敏感性。方法MTT法检测细胞生长情况;DAPI染色观察细胞核形态;电泳迁移实验(EMSA)检测NF-κB/p65与DNA顺式作川元什结合活性:Western Blot检测核蛋白中p65的含量。结果TSA预处理联合TRAIL作用肝癌细胞Be17402,MTT实验明显抑制细胞增殖;DAPI染色结果显示细胞核形态明显异常,可见浓染致密的颗粒荧光,典型细胞可见核固缩;EMSA结果表明NF-κB/p65与DNA结合带密度降低;Western结果显示细胞核蛋白中p65含量减少。结论TSA提高肝癌细胞Be17402对TRAIL的敏感性;TSA预处理联合TRAIL能通过降低p65的活性抑制细胞增殖和诱导细胞凋亡。
【Abstract】 Objective To investigate whether TSA could enhance the sensibility of Be17402 cells to TRAIL.Methods The growth station of cell is detected by MTT;The nuclear morphology was examined with DAPI staining;Binding activity of NF-κB/p65 with cis-acting elements of DNA was analyzed by electrophoretic mobility shift assay EMSA;The expression of p65 was detected by western blotting. Results Abnormal nuclear morphology was visible in cells treatment with recombined TRAIL and TSA,by fluorescence microscopy.The density of band of NF-κB/p65 DNA complex is lower in the Be17402 cells treatment with TRAIL and TSA.The content of p65 was decreased in the Be17402 cells treatment with TRAIL and TSA. Conclusion Squential treatment of Be17402 cells with TSA followed by TRAIL enhanced TRAIL cytotoxicity;TSA sensitized TRAIL-resistant Be17402 cells by downregulating the activity of p65,which could result in inhibition of cells proliferation and induction of cells apoptosis.
