全反式维甲酸通过抑制Src蛋白的表达诱导肝癌细胞BEL-7402凋亡

【摘要】 [目的]探讨全反式维甲酸(ATRA)诱导肝癌细胞凋亡及其影响ATRA作用的关键靶蛋白,为肝癌的治疗提供线索。[方法]不同浓度的ATRA处理人肝癌细胞BEL-7402和人正常肝细胞L-02后48小时,MTT法分析ATRA对细胞增殖的影响;并用DAPI染色法检测细胞凋亡情况,Western blot检测ATRA对细胞Src和p-Src蛋白表达的调控作用。[结果]ATRA对人肝癌BEL-7402细胞增殖有抑制作用,这种抑制作用呈剂量依赖性,随着药物浓度的逐渐增加,抑制作用呈递增趋势;2、全反式维甲酸(ATRA)能够诱导人肝癌细胞BEL-7402的凋亡,随药物浓度的增高,细胞凋亡率逐渐增高;3、全反式维甲酸(ATRA)对总Src蛋白的表达没有影响,可能通过抑制Src蛋白活性来诱导肝癌细胞BEL-7402的凋亡。
【Abstract】 Objective To explore the effect and mechanism of apoptosis induced by ATRA and identify several protein targets associated with the ATRA,which may provide clues for the treatment of liver cancer.Methods Human hepatoma cells BEL-7402 and normal human liver cells L-02 incubated with different concentrations of ATRA for 48 hours,The proliferation of cells was analyzed by MTT assay.The apoptosis was measured by DAPI staining and the expressions of Src and p-Src protein were detected by Western blot.Results 1,ATRA inhibited the proliferation of human hepatoma cell line BEL-7402 significantly in dose-dependent manners;2,The apoptosis of BEL-7402 cells was induced by ATRA;3,ATRA had no effect on the expression of total Src protein.It may be by inhibiting the activity of Src protein induced apoptosis ofhepatoceilular carcinoma cells BEL-7402.
