食物中毒及食品中金黄色葡萄球菌的肠毒素及基因的研究

【摘要】 目的:对食物中毒样品及食品中检出的金黄色葡萄球菌进行肠毒素基因分型,并采用mini-VIDAS法与其进行比较。方法:用PCR法测定金黄色葡萄球菌肠毒素基因(SEA-SEJ),用mini-VIDAS法测定肠毒素(SEA-SEE)。结果:从样品中分离的154株金黄色葡萄球菌菌株中,携带肠毒素基因SEA-SFJ的菌株为84.41%,同时携带2种及以上肠毒素基因的菌株占39.61%。在食品分离的菌株中,含传统的毒素基因型(SEA-SEE)的菌株占44.66%,含新发现的毒素基因(SEG-SEJ)的菌株占54.37%。在食物中毒标本中,含SEA-SEE基因的菌株占82.35%,含SEG-SEJ基因的菌株占41.18%。120株金黄色葡萄球菌菌株用mini-VIDAS法与PCR法分别测定肠毒素(SEA-SEE)及基因,结果显示完全符合的有96株,有24株结果不符合。结论:金黄色葡萄球菌食品分离株和食物中毒分离株在基因分布上有差异。普通PCR法测定肠毒素基因与mini-VIDAS法测定肠毒素结果上也有明显差异。
【Abstract】 Objective:To analysis the distribution of the enterotoxin genes of Staphylococcus aureus isolates between food poisoning and food samples.And the use of its mini-VIDAS method were compared.Methods:Staphylococcus aureus enterotoxin genes(SEA-SEJ) were determined by PCR and using mini-VIDAS determination of Staphylococcus aureus enterotoxin(SEA-SEE). Results:Isolated from the various types of samples of 154 strains of Staphylococcus aureus,84.41%of strains are carrying enterotoxin genes(SEA-SEJ),the isolates with more than one enterotoxin genes accounted for 39.61%.Strains isolated in the food contain the toxin genotype of traditional(SEA-SEE) accounted for 44.66%of the strains,the isolates with newly found enterotoxin SEG-SFJ genes accounted for 54.37%.In food poisoning,the isolates strains contain with SEA-SEE genes accounted for82.35%,the isolates with SEG-SEJ genes accounted for 41.18%.120 strains of Staphylococcus aureus were determined enterotoxins (SEA-SEE) and the gene by mini-VIDAS and PCR.The results showed that there is full compliance with the 96 strains,and there are 24 strains of the results do not meet.Conclusion:The distribution of enterotoxins genes are distinctly different between Staphylococcus aureus isolates from food and from food poisoning.Determination of ordinary PCR enterotoxin gene and mini-VIDAS method enterotoxin results have significant differences.
