8398甘蓝DH系组织培养和植株再生体系的初步研究

【摘要】 以甘蓝8398DH系子叶-子叶柄、下胚轴为外植体,在不同浓度6-BA与NAA激素组合的MS分化培养基诱导芽苗的分化,研究发现4-6天苗龄的8398DH系的子叶-子叶柄在MS+4.0 mg/L 6-BA+0.5 mg/L NAA+5 mg/L AgNO3的分化培养基中分化率为88.89%;下胚轴在MS+2.0 mg/L 6-BA+0.0 mg/L NAA+5 mg/L AgNO3的分化培养皿中分化率为98.11%。初步建立了甘蓝8398 DH系子叶子叶柄、下胚轴的高频率再生植株体系,为开展优良性状甘蓝新品种的生物技术育种工作提供了条件保证。
【Abstract】 In our experiment,the petiole with cotyledon and hypocotyl of Brassica oleracea"8398"DH line were taken as explant.Based on application of differentiation medium of 6-BA and NAA hormone combination with different concentration,we found that petiole with cotyledon of 4-6 days sterile seedling was suitable for differentiation Medium of MS+4.0 mg/L 6-BA+0.5 mg/L NAA+5 mg/L AgNO3,the regeneration frequency is 88.89%.For hypocotyl,the suitable differentiation Medium is MS+2.0 mg/L 6-BA+0.0 mg/L NAA+5 mg/L AgN03 with a 98.11% regeneration frequency.So,System of high frequency regeneration plant of Brassica oleracea was preliminarily established,which contributed good conditions to develop biological technique breeding work for good quality of Brassica oleracea.
