提高结球甘蓝游离小孢子胚诱导率的研究

【摘要】 13个甘蓝品种(品系)在NLN-13液体培养基中有10个品种诱导出胚。每花蕾产胚在0~1.8之间。比较NLN培养基中添加激素和活性炭的处理结果表明,NLN+13%蔗糖+活性炭0.5mg/L效果较好,胚诱导率提高3倍。接种后小孢子在33℃高温预处理24h产胚量较高。B5+6-BA0.2mg/L+3%蔗糖+1%琼脂培养基有利于小孢子胚长成植株。
【Abstract】 Isolated microspores from 13 varieties of rape were cultured in NLN-13 Liquid medium, 10varieties had produced microspore-derived embryoids.Embryo yield per bud was from 0 to 1.8. Hormone concentrations and active charcoal in NLN medium had been compared.NLN with 13% sucrose and active charcoal 0.5mg/L was better in production of microspore-derived embryoids. Average embryo yield was higher in 33℃high temperature treatment from 24 hour. Microspore-derived embryoids on B5 medium with 6-BA0.2mg/L and 3%Sucrose and 1%agar was good to regenerated plants.
