斑蝥素抗肿瘤作用的研究

【摘要】 在本研究中,首次在细胞水平上验证了斑蝥素及其衍生物的抗肿瘤作用。试验结果表明,斑蝥素及其衍生物对肿瘤细胞系有明显的抑制作用。斑蝥素、去甲斑蝥素和斑蝥素钠这三种药物对Hep3B细胞系的IC50值依次为:3.06 ug/ml、1.17 ug/ml和1.41 ug/ml;对Hela细胞系的IC50值依次为:9.52 ug/ml、6.36 ug/ml和8.58 ug/ml。Hela细胞的电镜照片显示,在经过斑蝥素处理1个小时后,其细胞结构发生了明显的变化,细胞内部许多超微结构被明显的破坏。在细胞核内出现了大量的深色的脂质小颗粒。细胞质中被诱导出了一些带状结构,出现了许多突出的空泡,还有一部分线粒体和内质网发生肿胀。结合电镜和实验结果,可以认为,斑蝥素导致肿瘤细胞急性死亡的主要原因是它削弱了肿瘤细胞线粒体的能量途径。

【Abstract】 Mylabris is the dried body of the Chinese blister beetle {Mylabris phalerata Pall.).The use of Mylabris can be traced back to more than 2,000 years ago in the traditional Chinese medicine.Cantharidin(hexahydro-3aα,7aα-dimethyl-4β,7β-epoxyisobenzofuran-l,3- dione) is the active ingredient of Mylabris.Mylabris is a potent vesicant and purported ar/hrodisiac used on human beings as well.In recent studies,Cantharidin was discovered to possess anti-tumor activities and would increase the number of leucocytes.In this research,we have verified its function of anti-tumor on the level of cells.The result show that cantharidin and its analogues have remarble effect on the tumor cell line.The IC50 number of these three activity agents toward Hep3B cells line are:3.06 ug/ml,1.17 ug/ml and 1.41 ug/ml,and the IC50 number of these three activity agents toward Hela cells line are:9.52 ug/ml,6.36 ug/ml and 8.58 ug/ml.The ultrastructured observations in the present study show that the structures of Hela cells treated with cantharidin for lh have changed.Hela cells show some lipid droplets,induced blebs and contained some swollen mitochondria.These findings suggest that cantharidin may be a potent cytotoxic drug for some cancer cells due to its ability,to impair the mitochondrial energy pathways.
