蛋白质组学方法筛选早期肺鳞癌相关蛋白

【摘要】 目的:应用蛋白质组学技术比较早期肺鳞癌组织和癌旁正常组织间的蛋白表达谱,以期发现早期肺癌癌变相关蛋白。方法:利用双向凝胶电泳技术分离人早期肺鳞癌组织和相应癌旁正常组织总蛋白,选择差异蛋白质点进行MALDI-TOF质谱分析和生物信息学分析。免疫组化技术验证部分差异蛋白质(ANXⅠ和HSP27)在肺鳞癌组织和癌旁正常组织中的差异表达。结果:肺鳞癌组织与癌旁正常组织的双向凝胶电泳图谱中平均蛋白质点数分别为626和602。选择在癌组织中高表达的10个差异蛋白点进行质谱分析,最终鉴定为ANXⅠ、HSP27等与细胞周期、信号转导、转录及翻译等功能相关的蛋白质。免疫组化方法检测结果显示ANXⅠ和HSP27在肺鳞癌组织中的阳性表达率均显著增高(P<0.05)。结论:蛋白质组学方法是一种用于初步筛选早期肺癌相关蛋白的有效方法,本研究所鉴定的早期肺鳞癌相关蛋白为进一步筛选用于肺癌的早期诊断及其治疗的分子标志奠定了基础。

【Abstract】 Objective:Searching the markers of lung cancer is the foundation to resolve the problem of early diagnosis.To compare the proteomic profile of early lung squamous cell carcinoma tissue and the adjacent normal tissue so as to find the early biomarker associated with lung carcinoma.Methods:The proteins of early lung squamous cell carcinoma tissue and adjacent normal tissue were separated by two-dimensional gel electrophoresis(2-DE) and differential proteins were screened to analyze by MALDI-TOF-MS.The part of differential proteins(ANXⅠand HSP27) were detected by immunohistochemical method to validate the reliability and accuracy of the results of mass spectrometry.Results:The proteomic profile of lung squamous cell carcinoma and the adjacent normal tissue was established and the average matched protein spots in the two group were 626 and 602 respectively.Ten protein spots which were screened withhigher expressional level in lung squamous cell carcinoma tissue were identified out by MALDI-TOF-MS as annexinⅠ(ANXⅠ)、heat shock protein 27(HSP27) and so on.The results of immunohistochemistry showed that the positive expression rate and strong positive rate of ANXⅠand HSP27 in lung squamous cell carcinoma tissues were all significant higher(P<0.05). Conclusion:Proteomic technology was an effective way for preliminary identification of the proteins associated with early lung carcinoma.The proteins which were identified in this study laid a solid foundation for screening the molecular markers of early diagnosis and treatment of lung cancer.
