两种测定双氟沙星与异育银鲫血浆蛋白结合率方法的比较

【摘要】 确立双氟沙星与异育银鲫血浆蛋白结合率的测定方法。考察了平衡透析法和超滤法分离游离药物浓度与总药物浓度的条件,并比较了平衡透析法和超滤法测定药物血浆蛋白结合率的差异。结果显示:透析袋与超滤膜对于药物的吸附率分别小于6.45±0.05%和4.51±0.08%;透析平衡时间为48h,超滤器在离心力12000g条件下离心120min药物分离完全;药物在血浆中和透析液(超滤液)中的回收率均大于92.5%和97.0%;日内精密度均小于0.99%和0.48%,日间精密度均小于0.84%和0.90%;平衡透析法与超滤法测定DIF血浆蛋白结合率分别为19.6-32.4%和19.24-30.01%。结果表明平衡透析法可以用于体外研究药物血浆蛋白结合性质,超滤法用于研究体内药物血浆蛋白结合性质。

【Abstract】 Established the method to assay the binding ratio of difloxacin(DIF) to plasma protein of Carassaisauratus gibebio,inspected the condition of equilibrium dialysis and ultrafiltration to seprate the free drug concentration and bond drug concentration,compared the plasma protein binding rate evaluate by HPLC(high performance liquid chromatography),equilibrium dialysis and ultrafiltration.The result showed that the adsorption of dialysis bag and ulrtafiltration membrane discern less than 6.45±0.051% and 4.51±0.077%;The equilibrium time of dialysis was 48h,the ultrafine filter completely separated in the centrifugal condition of 12000g,120min.The recovery rate of drug in blood plasma and dialysate(ultrafiltrate) were all higher than 92.5% and 97.0% respectively;The within-day precision were all less than 0.99% and 0.48%,inter-day precision all less than 0.84% and 0.90%;The binding rate of DIF by equilibrium dialysis and ultrafiltration was 19.6-32.4% and 19.24-30.01% respectively.The result showed the equilibrium dialysis can used to study drug plasma protein binding property in vitro,the ultrafiltration can used to study drug plasma protein binding character in vivo.
