化感物质肉桂酸乙酯对蛋白核小球藻生长及生理特性的影响

【摘要】 探讨了肉桂酸乙酯对蛋白核小球藻生长及生理特性的影响,从叶绿素a含量,抗氧化系酶活性,活性氧自由基(ROS)水平,丙二醛(MDA)含量以及光合活性的变化研究了其抑藻的生理生化机制。研究发现,肉桂酸乙酯对蛋白核小球藻具有快速抑制效应,随着浓度的增加抑制效果越明显。其96 h半效应浓度EC50值为5.45 mg·L-1。肉桂酸乙酯引起细胞内叶绿素a含量下降,活性氧自由基(ROS)的大量累积和MDA含量的增加。说明肉桂酸乙酯可能是通过引起活性氧自由基(ROS)的过度产生,或者由于肉桂酸乙酯的羟基与培养基中的过渡金属离子发生自氧化,产生H2O2,引起膜脂过氧化,造成细胞膜系统损伤,细胞功能发生紊乱。为了清除ROS,细胞通过提高抗氧化系酶的活性来保护其免受氧化损伤。肉桂酸乙酯对蛋白核小球藻细胞光合系统Ⅱ的最大光化学量子产量和潜在活性具有一定的急性抑制效应,但随着时间的延长藻细胞可以通过自身调节在一定程度上恢复其光合活性。肉桂酸乙酯具有易降解、挥发性、强抑制、效果快的优点,有望开发成生物除藻剂。

【Abstract】 The effects of ethyl cinnamate on the growth and physiological characteristic of Chlorella pyrenoidosa were studied.In order to explore the allelopathic mechanism,chlorophyll a content,the activities of antioxidant enzymes,reactive oxygen species(ROS) level, malondialdehyde(MDA) content and photosynthetic activity under ethyl cinnamate stress were also measured.The results revealed that ethyl cinnamate had acute inhibitory effects on the growth of Chlorella pyrenoidosa,and the inhibited degree tended to increase with increased concentrations.The effective concentration causing a 50%inhibition at 4 d was 5.45 mg ? L-l.Ethyl cinnamate could induce an obvious decrease in the contents of chlorophyll, over-accumulation of ROS and increase of MDA content Therefore,it suggested that the inhibitory effects might be due to the over-production of ROS or the generation of H202 induced by autoxidation between transition metal ions(Cu2+,Fe3+)in medium and phenolichydroxyl in the molecules of ethyl cinnamate,leading to lipid peroxidation and the damage of membrane system,ultimately interfering with the normal cellular metabolism.And for scavenging ROS,the algae cells were protected from oxidative damages through increasing the activity of antioxidant enzymes.The results demonstrated emyl cinnamate had acute inhibition to the maximum quantum yield and the potential activity of Chlorella pyrenoidosa, however,the photosynthetic activity could recover to some extent through their own regulation as rime goes on.Overall,ethyl cinnamate may have the potentiality of developing an algaecide due to its characteristic of degradability,volatility,strong and fast inhibition.
