念珠菌显色培养基的应用研究

【摘要】 念珠菌(Candida spp)是临床标本中最常见的酵母样真菌,也是重要的条件致病菌。传统的检验方法存在操作繁琐、检测周期长与特异性低等特点。本实验室研究开发出一种念珠菌显色培养基(HKMCC),与国外科玛嘉同类产品念珠菌显色培养基(CHROMagarCC)及传统培养基沙保氏琼脂(SDA),对相关菌株以及临床分离株在特异性、选择性及灵敏度方面进行了比较和初步评价。结果显示,HKMCC与CHROMagarCC显色培养基均具有较好的特异性,优于传统培养基SDA。HKMCC与CHROMagarCC显色培养基均具有较好的选择性,HKMCC对于非目标菌的抑制率比CHROMagarCC高9%,比传统培养基SDA高71.2%。8株念珠菌在三种培养基上的生长率均大于0.8。经统计分析,试验的三种培养基生长率差异不显著(P>0.05)。结论 :利用显色培养基检测念珠菌,特别是白色念珠菌,较常规的生化检测方法具有较高的特异性和选择性,具有实际较好的应用价值。
【Abstract】 Candida spp are the most frequently isolated yeast opportunistic pathogens which widely exist in clinical specimens.Because of the deficiencies of complicated operation and low specificity in the general method of the isolation and identification of Candida spp,a new chromogenic medium(HKMCC) was designed in this assay.Through detecting the reference and clinical isolated strains,the specificity,selectivity and sensitivity of HKMCC were studied and compared with CHROMagarCC and Sabouraud dextrose agar(SDA).The results showed that the specificity of HKMCC was as same as CHROMagarCC,and better than that of SDA. The selectivity of HKMCC was 9%higher than that of CHROMagarCC,and 71.2% higher than that of SDA for non target bacteria inhibition.The productivity of 8 strains on three culture media were all above 0.8,After statistical analysis of these results,there are no significant difference among three culture media(P > 0.05) in this study.In conclusion,Chromogenic Candida medium has a good prospect in applying,particularly C.albicans,because it has high specificity and selectivity.
