【摘要】 目的:建立wistar大鼠肾脏缺血再灌注损伤模型,探讨在大鼠肾脏近曲小管上皮细胞中寻找具有成体干/祖细胞特征细胞的可行性。方法:通过手术夹闭大鼠双侧肾动脉约50分钟后再次开放血流,以形成肾脏缺血再灌注损伤。于术后第3、5、7天取肾,免疫组化检测,观察在肾脏近曲小管上皮细胞中是否出现KIM-1、CD34、CD133、Brdu抗原表达及该细胞亚群的分布。结果:与阴性对照组相比,在实验组大鼠肾脏皮质中有KIM-1和CD133阳性细胞出现,而在皮髓质交界处和髓质中有CD34、Brdu阳性细胞出现。以上四种抗原的阳性细胞表达率随损伤后自我恢复时间的延长,而出现CD34及CD133抗原的阳性表达率在术后先上升然后再下降,Kim-1抗原的阳性表达率在术后下降,Brdu抗原的阳性表达率在术后呈现上升趋势。结论:在缺血再灌注损伤之后,肾脏皮质中表达KIM-1和CD133抗原的细胞明显增多,并且在肾脏髓质中表达CD34抗原和Brdu的细胞也明显增多,提示肾近曲小管上皮细胞发生损伤继发增殖再生,参与这一活动的细胞具有类似于上皮干/祖细胞极强的增殖分化修复损伤组织的能力,同时还具有表达CD133、CD34抗原的特征。这为探索肾脏成体干细胞研究及肾单位的组织重建可行性研究提供了依据。
【Abstract】 Objective By establishing a model of kindey ischemia/ reperfusion injury in Wistar rats,to study the feasibility about exploring of the cells with characteristics of stem/progenitor cells in proximal tubule epithelial.Methods The kidneys ischemia/reperfusion models were set up by blocking up the bilateral renal arteries about 50 minutes and recovering the kidneys’ perfusion of blood.Then the rats’ kidneys were taken off at the 3rd,5th,7th days of post-model.After undergone a series of treatment by a special process,immunohistochemistry staining was used to show the distribution and expression of KIM-1、Brdu antigen、CD34 and CD133 antigens in the kind of cells.Results Comparing to control group,the KIM-1 and CD133 antigens were showed up in cotex renis,meanwhile the CD34 and Brdu antigens were showed up in medulla renis.The expression rates of antigens rise or decline,which were according to the recovery time.(The number of cells that expressed CD34 and CD133 antigens rised in the firstly three days past operation, then it declined.Otherwise the number of cells that expressed Kim-1 antigens declined after operation.Meanwhile the expression mount of Brdu rises.) Conclusion Post-operation of ischima/reperfusion injurying in kidney,the expression rates of CD133 and KIM-1 antigen rised obviously in cotex renis,the same as the expression rates of CD34 and Brdu antigen in medulla renis.The result indicated that the phenomenon of proliferation appears in kidney proximal tubule.The cells that took part in the actions not only possessed of the strong proliferation and repairing ability of stem/progenitor cells,but also can expressed CD34 and CD133 antigen.It is the basis to research the tissue reconstruction of nephron and research in the field of kidney adult stem cell.
