改良制备异种脱蛋白骨免疫学研究

【摘要】 目的研究改良法制备的异种脱蛋白骨(heterogeneous bone,HGB)免疫学性质,为组织工程提供合适的支架材料。方法采用改良法制备猪脱蛋白骨,将浸提液进行小鼠安全性研究及修复山羊胫骨大段骨缺损,进行免疫学及成骨系列试验。结果小鼠淋巴细胞增殖试验、外周血及植骨局部CD4~+、CD8~+淋巴细胞亚群,血清特异性抗体IgG检测,细胞因子检测在各时相点明显低于新鲜猪骨。大动物在体免疫学及成骨试验与自体骨相当。结论改良法制备猪脱蛋白骨符合医用生物材料应用要求,组织相容性好,安全无毒。不引起明显的细胞和体液免疫反应,有良好的组织相容性。异种DPB不影响自体MSCS成骨,与rhBMP2复合物成骨能力与自体骨组相当,可作为组织工程支架材料修复大动物长骨大段缺损,新骨生物力学与自体骨组相当。改良法法制备异种DPB可作为骨组织工程支架材料。

【Abstract】 Objective to research the immunological properties of dissimilar deproteinization bone prepared by modified method and provide a suitable scaffold for tissue engineering.Methods prepared deproteinization pig bone by modified method,and the water extract was used in the safety studies,and repaired the segmental bone defect of goat tibia,and did immunological and osteogenic serials tests.Results mice lymphocyte proliferation test,CD4 + and CD8 + lymphocyte subsets in peripheral blood and at the site of implantation,serum specific antibody IgG detection and cytokine test showed significantly lower results than those of fresh pig bones at each time point.The large animals’ results of in vivo immunological test and osteogenic test were similar to the results of autologous bone.Conclusion the deproteinization pig bone prepared by modified method is corresponding with the application requests for medical biological materials,with excellent histocompatibility and innocuity safety.No obvious cellular and humoral immune response and had good histocompatibility.And not affected MSCs osteogenisis of autologous bone.And the osteogenic ability of heterologous DPB compounded with rhBMP2 was rather to this of autologous bone,which could be used as a suitable scaffold for tissue engineering to repair segmental bone defect of large animals and the newborn bones have similar biomechanixs with autologous bones.Therefore,it could be used as scaffolds for bone tissue engineering.
