汉防已甲素对大鼠急性脊髓损伤后神经细胞凋亡的影响

【摘要】 目的动态观察大鼠急性脊髓损伤后神经细胞凋亡情况及汉防已甲素(Tetrandrine,Tet)对其的影响,为临床上治疗急性脊髓损伤提供实验依据。方法选用95只成年Sprague-Dawley(SD)大鼠,随机分为正常对照组,损伤组,A组、B组。改良Allen’s重物打击法制作脊髓损伤模型。在损伤局部,损伤组滴入生理盐水,A组滴入Tet,B组滴入甲基强的松龙,A组和B组动物术后3d每日腹腔分别注射Tet和甲强龙。对照组于术后21d处死,余三组分别于术后8h,1d,3d,7d,14d,21d处死,取损伤段脊髓5mm,上段石蜡切片HE染色,下段用生理盐水浸泡,零下20℃低温保存用作流式细胞仪半定量检测凋亡细胞。结果1组织学检测组织损伤情况A组优于损伤组,B组优于A组,术后8h~21d动态观察,3~7d达损伤高峰。2凋亡细胞半定量分析各组均在损伤后7d达到凋亡细胞数峰值,8 h~7 d逐渐增多,7 d-~21 d有轻度减少趋势。同一时点组间比较A组明显少于损伤组,B组少于A组。结论急性脊髓损伤后7 d达到神经细胞凋亡高峰,Tet有效抑制了神经细胞凋亡,保护了脊髓功能。

【Abstract】 Objective To observe the mechanism of neuronal apoptosis and prefective effect of Tetrandrine(Tet) after acute spinal cord injury(ASCI) in SD rats.To provide the possibility of treatment for ASCI.Methods Ninety - five Adult SD rats were divided at random into four groups:control group(5),ASCI group(30),group A(30),group B(30).The animal ASCI model was established with Allen’s method.The rats in control group were killed at 3 weeks later.The rats of the other three groups were sacrificed at 8 hours and 1,3,5,7,14,21 days after surgery,the spinal cord tissue of 5mm long were taken out and divided into two parts,one part tissue sections was stained with hematoxylin and eosin and the spinal cord tissue was observed,another part tissue was put into the flow cytometry(FCM).The scan quantity analysis for apoptosis nerve cells can be gained. Results(1)The injury of spinal cord tissue is most seriously in SCI group,group A is better than it and group B is better than group A.Observed from 8h to 21d after operation,the injury reach the top during 3-7 days.(2)The number of apoptosis neurons in SCI group was more than that in group A,and group A was more than that in group B.The number reached the top at 7 days later. conclusion Apoptosis was the main pattern by which the nerve cells died in ASCI and Tetrandrine could inhibit apoptosis
