BMP2、VEGF165双基因转染小鼠骨髓基质干细胞的体内诱导成骨

【摘要】 目的:观察骨形成蛋白2(BMP2)、血管内皮生长因子165(VEGF165)双基因转染的小鼠骨髓基质千细胞是否具有异位诱导成骨能力。方法:脂质体介导下将双基因真核表达质粒pIRES-BMP2-VEGF165导入小鼠骨髓基质干细胞。用RT-PCR和免疫组织化学方法观察BMP2和VEGF165双基因在小鼠骨髓基质干细胞内的表达;将转染BMP2和VEGF165双基因的小鼠骨髓基质干细胞植入4只裸鼠大腿后群肌袋内,4周后用影像学和组织学方法观察体内诱导成骨作用。结果:转染BMP2、VEGF165的小鼠骨髓基质干细胞有明显的BMP2和VEGF165 mRNA及其蛋白表达,此细胞植入裸鼠大腿后群肌袋,4周后有大量异位骨形成。结论:转染BMP2和VEGF165双基因的小鼠骨髓基质干细胞具有较强诱导异位成骨能力。

【Abstract】 Objective To evaluate the osteogenic potential efficacy of mouse bone marrow stromal cells(mMSCs) transfected with bone morphogenetic protein 2(BMP2) and vascular endothelial growth factor165(VEGF165) genes. Methods The mMSCs were transfected with pIRES - BMP2 - VEGF165 by liposome - mediated method.The co - expression of BMP2 and VEGF165 genes were determined by RT - PCR and immunohistochemical analysis.The transfected cells were injected into the thigh muscle pouches of four nude mice.The osteoinductivity of the transduced cells were evaluated by radiography and histology analysis in 4 weeks after injection. Results RT- PCR and immunohistochemical analysis showed that these transfected cells indeed could produce the mRNA and proteins of the BMP2 and VEGF165.The ectopic bones formed in the thigh muscle pouches of the nude mice were obviously observed 4 weeks after cell injection. Conclusion The mMSCs transfected with BMP2 and VEGF165 genes can induce ectopic osteogenesis of mouse marrow stromal cells.
