高效液相色谱法测定人血浆中5-FU的浓度

【摘要】 目的:建立用高效液相色谱法测定人血浆中5-FU的浓度。方法:采用UltimateC18色谱柱(250mm×4.5mm,5μm);流动相:10mmol·L-1KH2PO4缓冲溶液;流速:1.0mL·min-1;检测波长:265nm;柱温:40℃。结果:5-FU在0.48μg·mL-1~61.4μg·mL-1范围内线性关系良好(r=0.998),最低检测浓度0.48μg·m-1,日内和日间RSD分别为5.12%~9.98%(n=5)和5.28%~10.95%(n=5)。室温放置,冷冻保存,反复冻融条件下稳定性良好。结论:该法具有操作简便可靠,准确,稳定性高等特点,适合应用于人血浆中5-FU的浓度测定。

【Abstract】 OBJECTIVE:To develop an HPLC method for the determination of 5-fluorouracil concentration in human plasma.METHODS:The concentration of 5-fluorouracil was assayed on a Ultimate C18 column with a mobile phase consisting of 10mmolL-1KH2PO4 at a flow rate of l.0mL.min-1 and detected at 265nm.RESULTS:The calibration curve of 5-fluorouracil was linear over the range of 0.48μg.mL-1~61.4μg.mL-1 (r= 1.0000).The low limit of determination was 0.48μg.mL-1.The RSD of the intra-day and inter-day precisions were 5.12%9.98%and 5.28%~10.95%respectively.The samples were stable in room and low temperature,and were also stable in Free and thaw condition.CONCLUSION:This method is simple,precise,stabile and suit to be applied in the determination of 5-fluorouracil concentration and its Pharmacokinetic studies.
