香椿乙醇提取物对糖尿病模型小鼠降血糖作用的初步研究

【摘要】 目的探导香椿乙醇提取物对糖尿病模型小鼠的降血糖作用及机理。方法把四氧嘧啶用0.9%NS(生理盐水)配成0.02g.mL-1,按200mg.kg-1注射到SPF(无特定病原体)级小鼠的腹腔,5天后用葡萄糖氧化酶法测血糖,空腹血糖〉10.00mmoL.L-1为选模成功小鼠。将小鼠随机分为4组各10只,阴性对照组(0.9%NS)、阳性对照组(二甲双胍70mg.kg-1)、香椿醇提物高剂量组(1g.kg-1)、低剂量组(0.5g.kg-1);另设正常组10只(蒸馏水),均每天灌胃一次,连续7天,末次给药后禁食24h,然后空腹眼眶静脉取血,用葡萄糖氧化酶法测血糖,用放免法测血清Insulin(胰岛素)。结果香椿乙醇提取物高、低剂量组与生理盐水组(模型组)比较,血糖水平明显降低P〈0.05,**P〈0.01;血清胰岛素水平明显增高#P〈0.05,##P〈0.01,均具有显著性差异。结论香椿乙醇提取物对四氧嘧啶所致的高血糖模型小鼠有显著的降血糖作用,其作用机理可能与升高血清胰岛素水平有关。

【Abstract】 OBJECTIVE:To investigate the effect and mechanism of alcohol extracts from Toon sinensis on reduce blood sugar of alloxan diabetic rats. METHODS:To confect alloxan 0.02g/ml with 0.9%NS and injected alloxan into abdominal cavity to inducediabetic model,and the blood glucose was detected by glucose oxidizes after 5 days.When hypoglycemic point>10.00m mol/L,the rats was asuccessful model made.40 SPF model rats were divided into 0.9%NS negative contral group,metformin positive contral group(70mg/kg),high and low doses of alcohol extracts from Toon sinensis(1g/kg , 0.5g/kg), 10 rats each group and in addition make a normal contral group of 10 healthy rats,all groups were fed tid for 7 consecutive days and 24 hours forbid to feed after finaly meal,then sample the blood pass eye socket on empty stomach,and the blood sugar and insulin were detected by glucose oxidizes and radioimmunoassay .RESULTS: The blood sugar and insulin of the high and low does of alcohol from Toon sinensis is showed significant differences as against the 0.9%NS group.CONCLUSION:The alcohol extracts from Toon sinesis could effectively decrease the blood sugar of alloxan diabetic rats and the mechanism of action to hoist the level of insulin.
