菲律宾蛤仔糖胺聚糖RG-1的结构特征及免疫活性研究

【摘要】 目的:对菲律宾蛤仔糖胺聚糖RG-1的一级结构特征及细胞免疫调节活性进行研究。方法:采用高效液相色谱法测定分子量和单糖组成,红外光谱、高碘酸氧化和Smith降解等研究其糖链主要结构特征;MTT法分析对小鼠脾淋巴细胞的影响。结果:R G-1主要由葡萄糖、氨基半乳糖和半乳糖通过1→6、1→4或1→4,6键连接构成主链,在主链中还含有大量的半乳糖醛酸,重均分子量为9.94×10~5。RG-1显著增强小鼠腹腔巨噬细胞吞噬中性红的作用和脾淋巴细胞的增殖(p<0.01)。结论:菲律宾蛤仔糖胺聚糖可通过增强细胞免疫而起到抗肿瘤作用。
【Abstract】 Objective:To investigate the structural fecture and immunomodulatory activity of the glycosaminoglycan RG-1 from Ruditapes philippinarum.Methods:The chemical structure of the glycosaminoglycan RG-1 was elucidated by HPGPC,IR,periodate oxidation and Smith degradation;Effects of RG-1 on proliferation of KM mouse spleen lymphocytes were detected by MTT.Results: RG-1 was composed of Glc-GalNAc-Gal by 1→6,1→4 or 1→4,6 in main chain backbone with Mr 9.94×10~5.RG-1 significantly simulated the phagocytic activity of mice peritoneal macrophages and proliferation of KM mouse spleen lymphocytes in vitro (p<0.01).CONCLUSION Glycosaminoglycan RG-1 can inhibit the cancer by enhance cell immunomodulatory activity.
