低聚糖阿魏酸酯含量的快速测定研究

【摘要】 低聚糖阿魏酸酯是以纤维质为原料获得的一类新型食品功能因子,具有良好的抗氧化活性和益生菌增殖功能,但由于低聚糖聚合度和阿魏酸结合量不同而导致检测困难。本研究分别以麦麸、玉米芯、玉米麸皮为原料,采用酶法制备低聚糖阿魏酸酯,探讨了其快速检测方法,并通过高效液相色谱法验证了该法的准确性。结果表明,采用Amberlite树脂分离纯化可除去样品中的低聚糖(不含结合态的阿魏酸)、双波长分光光度法(345 nm,375 nm)结合还原糖法可用于对低聚糖阿魏酸酯进行准确定量。

【Abstract】 Feruloylated oligosaccharides,prepared from plant cell wall materials,are new functional ingredients which showed high antioxidant and prebiotic activity.However,it is difficult to determine them because of their different degree of polymerization of the sugars between prepared samples and different amount of bound ferulic acid between the raw materials.In this research,feruloylated oligosaccharides were respectively prepared from wheat bran,corn cob,corn bran by enzymic hydrolysis and their rapid determination was investigated,while the veracity of the method was verified by HPLC.The results showed that,after purification by Amberlite resin chromatography to remove the non-ferulic acid bound oligosaccharides,the DW-spectrophotometry(345nm,375nm) could effectively determine the content of ferulic acid bound to the oligosaccharides;Combined with the determination of reducing sugars after acidolysis, it could be precisely quantitatively determine the total content of feruloylated oligosaccharides,the content of the bound ferulic acid and oligosacchrides respectively.
