靶向SERS探针的制备及其活细胞中成像中的应用
Preparation of Nuclear and Membrane Targeting SERS Nanoprobes for Simultaneously Raman Imaging in Live HeLa Cells

【摘要】 通过柠檬酸钠还原法制备金纳米颗粒作为增强基底,叶酸和核定位肽作为靶向分子,并且使用对琉基苯甲酸和甲酚紫作为信号分子,分别制备出膜靶向探针和核靶向探针。将设计的探针与细胞共培养后使用激光共聚焦拉曼显微镜进行检测,分别在细胞核和细胞膜检测到相应的信号分子的特征拉曼位移,并以此进行细胞成像的绘制,清晰地呈现出细胞的轮廓和细胞核的位置。稳定高灵敏的细胞靶向SERS探针在生物诊断、病理检测和药物靶向释放等技术领域将产生深远影响。
【Abstract】 Herein two kinds of surface-enhanced Raman scattering(SERS) nanoprobes were prepared for simultaneously imaging of the nucleus and the cell membrane.Folic acid(FA) and SV-40 large T nuclear localization signal(NLS) peptide attached to the gold nanoparticles for targeting cell membrane and nucleus of specific canner cells.At the same time,sulfanylbenzoic acid(4-MBA) and cresyl violet acetate attached to the GNP as Raman reporter molecules.Raman signals from both 4-MBA and cresyl violet acetate has been detected on cell membrane and nucleus,respectively.Raman mapping technique is capable of describing the location of these two probes in single cells.
