光谱法表征纳米金与溶菌酶之间的相互作用
Characterization of Interactions between Gold Nanoparticle and Lysozyme by Spectral Methods

【摘要】 研究纳米粒子与蛋白质的相互作用对了解纳米粒子的环境效应和生物效应具有重要的意义。采用紫外-可见光谱法、荧光光谱法和圆二色谱法对纳米金与溶菌酶之间的相互作用进行了表征。实验结果表明,溶菌酶可引起纳米金的聚集,而纳米金可以诱导溶菌酶分子中α-螺旋含量减少;荧光淬灭表明纳米金与溶菌酶之间的结合常数为2.9×1011L·mol-1;此外,温度和加样顺序等条件对纳米金和溶菌酶之间相互作用有很大的影响;高浓度的NaCl和阳离子表面活性剂十二烷基三甲基溴化铵(DTAB)能够引起纳米金的严重聚集,而溶菌酶的存在则可以明显抑制此类聚集。
【Abstract】 It is of great importance for understanding environmental and biological effects of nanoparticles to study interactions between nanoparticles and proteins.The interactions between gold nanoparticles(AuNPs) and lysozyme were characterized by UV-Vis absorption,fluorescence and circular dichroism(CD).The results showed lysozyme could cause drastic aggregation of AuNPs,while AuNPs could induce decrease ofα-helix in the secondary structure of lysozyme;Fluorescence quench indicated that binding constant between lysozyme and AuNPs was 2.9×1011 L ? mol-1;Additionally,temperature and sample introducing order had great effect on the interactions between AuNPs and lysozyme;NaCl with high concentration and cationic surfactant dodecyltrimethylammonium bromide(DTAB) could cause serious aggregation of AuNPs,while lysozyme could obviously inhibit the aggregation.
