正己烷和聚乙二醇对水溶液中溶菌酶构象影响的红外光谱分析

【摘要】 运用傅里叶变换红外光谱法(FTIR)研究了正己烷、聚乙二醇对水溶液中溶菌酶(Lyz)构象的影响。结果表明,聚乙二醇-200与溶菌酶水溶液混合后,溶菌酶的构象无明显变化;而正己烷与溶菌酶溶液混合后,溶菌酶酰胺I带中的反平行β-折叠或/和β-转角含量增多。从而印证了我们关于表面的亲水、疏水性质对吸附蛋白质构象影响的结论。
【Abstract】 The effect of n-hexane and polyethylene glycol on the conformation of lysozyme in aqueous solution was investigated by FTIR spectroscopic analysis.No significant conformational change of lysozyme was found after mixing hydrophilic poly(ethylene glycol) with lysozyme solution.However,the amount of anti-parallel P-sheet and/or P-turn structures of lysozyme in the amide I region increased after blending hydrophobic n-hexane with lysozyme solution.The conformational changes of lysozyme adsorbed on hydrophilic and hydrophobic surfaces were well confirmed in this study.
