唐古特红景天(Rhodiola tangutica)有效化学成分及红景天苷含量的研究

【摘要】 本文采用超声波法分别用正丁醇、乙醇、乙酸乙酯、二氯甲烷、四氯化碳5种不同极性的溶剂对唐古特红景天(Rhodiola tangutica)有效成分进行提取,并将萃取液浓缩用0.45 um的有机滤膜过滤后经毛细管气相色谱-质谱联用法(GC-MS)分离鉴定并按峰面积归一化法计算相对含量。共鉴定出64种成分,主要为醇类(24.58%)、黄酮类(8.96%)、有机酸类(8.45%)、酯类(5.71%)和烷烃类(14.78%),其中相对含量较高的成分有己醇(7.34%)、丁醇(4.57%)、2-丁酮(1.79%)、3-甲基-2-(H)-呋喃酮(1.87%)、二十四基乙酸(1.77%)、1,2-苯二羟酸异辛酯(0.97%)、十三烷(2.43%)、十一烷(2.47%)等。以红景天根茎粉末为原料,经70%乙醇溶剂连续回流提取,采用硅胶柱色谱层析进行分离纯化,利用高效液相色谱法(HPLC)测定出大花红景天(Rhodiola crenulata)中红景天苷含量为0.92%,唐古特红景天中红景天苷的含量为1.20%。唐古特红景天中红景天苷含量高于大花红景天中红景天苷的含量。
【Abstract】 This paper describes the extraction procedure of Rhodiola tangutica using the ultrasonic method with 5 different solvents:n-butanol,ethanol,ethyl acetate,dichloromethane,and carbon tetrafluoride.Resultant concentrates are then filtered with 0.45 micrometer organic membrane filters and finally separated and identified using the GC-MS method.Upon analysis of the concentrates’ relative content using peak area normalization,result is as follows:64 compounds were identified,including various alcohols(24.58%),flavonoids(8.96%),organic acids(8.45%),esters(5.71%) and alkanes(14.78%).More specifically,major compounds are:hexanol(7.34%),butanol(4.57%),2-butanone(1.79%),1-methyl-2-(H)-furanone(1.87%),tetracosyl ethanoic acid(1.77%),isooctyl 1,2-dihydroxybenzoate(0.97%),tridecane(2.43%),undecane(2.47%),etc.To determine the concentration of salidroside in Rhodiola algida var.We performed continuous reflux extraction on its rhizome powder with 70%ethanol solution,conducted separation and purification through silica gel column chromatography,and applied highperformance liquid chromatography.The salidroside content of Rhodiola crenulata is 0.92%,The salidroside content Rhodiola Tangutica is 1.20%.The salidroside content of Rhodiola Tangutica is higher than that is in Rhodiola crenulata.
