预调制可储存性根管充填糊剂的研制
A new preservable calcium phosphate-based root canal sealer

【摘要】 磷酸钙骨水泥作为一种新型的根管充填材料,以其生物相容性高和易塑形的特点得到材料界和医学界的广泛关注。用液相后移植工艺方法合成钙稳定介孔氧化锆,将甲硝唑负载其孔道内,作为根管消炎显影剂。在骨水泥中加入一定含量的保湿剂、增稠剂以及抗菌消炎显影剂,制得可存储性根管充填材料。结果表明,添加剂的加入增强了糊剂的稳定性,当糊剂保存在4℃,50%相对湿度条件下,两个月内糊剂的物化性能和显微结构几乎没有变化。该糊剂具有良好的显影性及生物相容性,甲硝唑在一段时间内可以缓慢释放达到根管消炎的作用。
【Abstract】 Preservable root canal sealer was considered as a promising material for the therapy of pulpitis and periapical periodontitis.The objective of this study was to develop a new kind of preservable root canal sealer based on calcium phosphate cement(CPC).The sealer consisted of calcium phosphorite powers,gelling agent and moist agent.Metronidazole-loaded mesoporous calcium-stabilized zirconia,as a developer,was added to increase the sterilizing effect.Its physicochemical properties and biological properties were measured.The physi-cochemical properties of sealer had no obvious change during two months when premixed and stored at 4℃,50% relative humidity(RH).The sealer exhibited good radiopacity and sealing ability.Agar dilution was used for determining antibacterial activity in vitro of it,when 5wt%metronidazole was added,the minimum inhibitory concentration(MIC) of Streptococcus mutants was 5mg/mL and others were all less than 0.156-0.312mg/mL. In the following eight days,the metronidazole could be released slowly so as to inhibit the growth of bacteria. The results of MTT showed the cell survival rate was up to 100%.With the addition of moist agent and gelling agent,the workability and the microstructure of sealer hardly changed in two months after being stored at 4℃, 50%RH.When it was injected into root canals,it could cure in 45~55h and be gradually transformed into non-stoichiometric hydroxyapatite which was similar to the apatitic mineral found in teeth.
